Objective: The chickpea dietary fiber developed from chickpea skin in Xinjiang was used as an intervention,to observe its effects on lipid metabolism and intestinal microflora in hyperlipidemia rats induced by high-fat diet,and to preliminarily explore the regulatory effect of chickpea dietary fiber on blood lipid and intestinal microflora in hyperlipidemia rats.Methods:(1)Establishment of hyperlipidemia model and group intervention: Ten of50 male SD rats were randomly selected as normal control group and fed with basic diet,the rest were used as hyperlipidemia model and fed with high-fat diet.After 7 weeks,the levels of TC and TG were detected,the significant increase of TC and TG after modeling was regarded as the success of the establishment of the model(p<0.05).The 40 hyperlipidemia model of rats were randomly divided into hyperlipidemia model group,chickpea fiber high(30 g/kg high-fat diet),middle(15 g/kg high-fat diet),and low(5 g/kg high-fat diet)three intervention groups,10 in each group,and intervention groups were given high-fat diet containing different levels of chickpea dietary fiber.(2)Sample collection and index detection: After 7 weeks of intervention,animals were treated,abdominal aorta blood was collected for 3 ml,and serum lipid and oxidative stress levels were measured;The liver and epididymis fat were dissected and the liver weight and periepididymis fat weight were measured of rats in each group;feces were taken aseptically for genome 16 S rDNA sequencing and fecal short-chain fatty acids content detection.Results:(1)Compared with the hyperlipidemia model group,the weight gain,liver weight and epididymal lipid weight were decreased in Chickpea dietary fiber group.(2)Blood lipid levels: Compared with the hyperlipidemia model group,the TC,TG,LDL-C values of the chickpea dietary fiber group were significantly lower(p<0.05),and the HDL-C values were significantly increased(p<0.01).(3)Antioxidant capacity:Compared with the hyperlipidemia model group,in each dose of chickpea dietary fiber,the MDA content decreased(p<0.05),and the activity of T-SOD and GSH-PX increased(p<0.05).(4)Sample diversity analysis: Alpha diversity analysis showed that the diversity of intestinal flora in the normal control group and the chickpea dietary fiber group was higher than that in the hyperlipidemia model group.Beta diversity analysis showed that there were significant differences in flora composition among hyperlipidemia model group,normal control group and intervention group.Rats in each intervention group tended to be similar and interactive.(5)Intestinal flora composition analysis: high-fat diet can increase the number of Firmicutes in rats,reduce the number of Bacteroides,Lactobacillus and Bacillus,chickpea dietary fiber intervention group can effectively increase the contents of Bacteroides,Lactobacillus and Bacillus in rats.(6)Fecal short-chain fatty acids: The content of fecal short-chain fatty acids in hyperlipidemia model group decreased,and the level of propionic acid decreased significantly(p<0.05).The content of fecal short-chain fatty acids in each intervention group increased in varying degrees,and the effect of high-dose group was the most obvious.Conclusion: Chickpea dietary fiber can reduce the body weight of hyperlipidemia rats and regulate abnormal blood lipids,its lipid-lowering effect may be related to raising the antioxidant capacity,improving the diversity and richness of intestinal flora,increasing the proportion of beneficial flora and regulating the content of short-chain fatty acids in hyperlipidemia rats.
