Objective:To investigate the effect of Yiqi Jianpi Huayu recipe combined with paclitaxel liposome metronomic chemotherapy on the expression of epithelial-mesenchymal-transformation(EMT)markers and related molecules in the lung metastasis model of MFC cells in gastric cancer mice,so as to elucidate the mechanism of affecting the invasion and metastasis of gastric cancer.Experimental methods:1.Fifty mice were randomly divided into 4 groups.namely model group,traditional Chinese medicine(Yiqi Jianpi Huanyu recipe,YQJPHY 20g/kg)group,paclitaxel liposome(5mg/kg)group and(YQJPHY+ paclitaxel liposome)combined group,with 10 rats in each group.Injecting MFC cells into the subcutaneous tissue and tail vein of mice.,was performed to establish the mice gastric carcinoma models and experimental lung matastasis models,respectively.The growth of mice was observed every day and weighed every 3 days.After the last administration,the lung,spleen and thymus were stripped completely,and the changes in growth,body mass and body before and after administration were compared.The number of lung surface metastases,inhibition rate of lung metastasis,thymus index and spleen index were calculated.2.RT-PCR was used to detect the effect of Yiqi Jianpi Huanyu recipe comb.ined with paclitaxel liposome metronomic chemotherapy on EMT-related genes(E-cad,N-cad,Slug,Snail)in lung tissues of MFC cell lung metastasis model 615 mice.3.Western Blot was used to detect the effect of Yiqi Jianpi Huanyu recipe combined with paclitaxel liposomal metronomic chemotherapy on EMT-related proteins(E-cad,N-cad,Vimentin,Slug,Snail)in lung tissues of MFC cell lung metastasis model 615 mice.Results:1.Compared with the model group,the body mass of mice in the YQJPHY group was significantly increased(P<0.01),the body mass of mice in the paclitaxel liposome group was slightly increased(P<0.05),and the body mass of mice in the combined group was significantly increased(P<0.01).At the same time,the mental state of mice in the combined group was significantly better than that in the paclitaxel liposome group,with black and bright fur and more flexible activities.2.Compared with the model group,the number of lung surface metastases was significantly reduced in the YQJPHY group,the paclitaxel liposome group and the combined group(P<0.01).Compared with the paclitaxel liposome group,the number of lung surface metastases was significantly increased in the YQJPHY group(P<0.01),while the number of lung surface metastases was significantly reduced in the combined group(P<0.01).Both paclitaxel liposome and Yiqi Jianpi Huayu recipe can significantly reduce the number of surface metastases,while the combination of-paclitaxel liposome and Yiqi Jianpi Huayu recipe has a better inhibitory effect on metastasis.The inhibition rates of lung metastasis in the YQJPHY group,the paclitaxel liposome group and the combined group were 11.76%,47.84%and 66.27%.This indicated that paclitaxel liposomes could inhibit lung metastasis in mice,and the inhibition rate of lung metastasis in mice was significantly increased after the combination of Yiqi Jianpi Huanyu recipe.3.Compared with the model group,thymus index and spleen index were increased in the YQJPHY group(P<0.05).The thymus of some mice in the paclitaxel liposome group was not visible,and the spleen was significantly reduced.Compared with the model group,the thymus index and spleen index of mice in the paclitaxel liposome group were decreased(P<0.05).Thymus and spleen tissues were observed in mice of the combined group.Compared with the model group,the thymus index and spleen index of mice in the combined group were increased(P<0.05).Compared with paclitaxel liposome group,thymus index and spleen index were significantly increased in the YQJPHY group(P<0.01),and thymus index and spleen index were significantly increased in the combined group(P<0.01).This indicates that paclitaxel liposome can damage the immune function of mice,but the combination of Yiqi Jianpi Huayu recipe can alleviate the damage.4.Through RT-PCR and Western blot experiments,EMT-related genes and proteins in the lung tissues of mice were detected,and it was found that paclitaxel liposome combined with Yiqi Jianpi Huayu recipe could regulate the expression of EMT-related genes and proteinsConclusion:Paclitaxel liposomes can inhibit the metastasis of gastric cancer to the lung,and can better inhibit the progress of metastasis the combination with Yiqi Jianpi Huayu recipe.The mechanism may be related to the up-regulation of E-cadherin expression and down-regulation of N-cadherin,Vimentin,Slug and Snail expression. |
