Objective:(1)To explore the improvement of fagopyrum cymosum(Trev.)meisn(FAG)tablets on DSS-induced inflammatory bowel disease in mice;(2)To explore the differences between WBC,CRP,ESR,et al during the active and remission period of IBD.Methods:(1)20 SPF C57BL/6 mice were randomly divided into model group and FAG group.The mice in both groups were fed 3%DSS for 7 days,with model group administrated double distilled water and FAG group administrated FAG suspension once a day for 7 days.Simultaneously record daily mouse body weight,stool viscosity,bloody stool were and then calculate disease activity index.After 7 days,and then fasted for 24 hours,the mice were sacrificed,dissected and drew the material.The length of the intestine and spleen weight from these mice were measured and the spleen index was calculated.The sections of the colorectal tissue with HE staining were followed by histopathological scor:ing.The relative expression levels of TNF-a,IL-6,IL-1β,IL-10 and ZO-1 and Occludin were detected by real-time PCR,and the content of serum endotoxin was detected by tachypleus amebocyte lysate,and then,proportion of proliferating cells in IECs was detected by Ki67 immunohistochemistry;(2)From 143 cases of IBD admitted to Nantong Third People’s Hospital from January 01,2014 to August 31,2018,94 eligible cases were screened out.By the improved Mayo scoring system and simplified CDAI calculation,these 94 cases were divided into active and remission groups,and then the differences of WBC,NEUT%,HGB,PLT,EO,HCT,PDW,CRP,CRPH and ESR were compared between these two groups.Results:(1)Compared with the model group,the DAI scores of the FAG group were significantly decreased on the 5th and 6th day(P<0.01),the length of the colorectal was significantly increased(P<0.05)and the spleen index was significantly decreased(P<0.001).And then,the histological score was significantly lower(P<0.05),and the expression level of TNF-α,IL-6 and IL-β in the colorectal issues was significantly down-regulated(P<0.05),and the content of serum endotoxin was significantly reduced(P<0.05).Besides,FAG can promote the proliferation of impaired IECs(P<0.05);(2)In 94 cases of IBD,PLT,CRP,CRPH and ESR was significantly increased(P<0.05)and PDW was significantly decreased(P<0.05)compared with the remission period.Conclusion:(1)FAG can preventively alleviate the intestinal and systemic symptoms of IBD,reduce the inflammatory damage of colorectal tissue and systemic inflammatory response by down-regulating the expression of pro-inflammatory factors,and promote proliferation of impaired IECs for intestinal mucosal healing;(2)The active and remission periods of patients with IBD can be distinguished by examining PLT,PDW,CRP,CRPH and ESR in the blood,which provides ideas for subsequent clinical use.
