Objective: To prepare targeted ultrasound contrast agent(MBt)with CXCL12 antibody,and to investigate the correlation between the parameters of targeted ultra sound contrast agent and MVD of ovarian cancer transplantation models at differen t stages.Methods: Sixty nude mice with ovarian cancer transplanted tumors were r andomly divided into 6 groups(10d,20 d,30d,40 d,50d and 60 d).CXCL12 antib ody-targeted ultrasound contrast agents were prepared.The normal(MBc)and targe ted contrast-enhanced ultrasound(MBt)examinations were performed successively t o obtain time-intensity curve(TIC)of contrast-enhanced ultrasound(CEUS).The ne ovascularization density(MVD)of nude mice tumors was counted by CD31 immuno histochemistry.The correlation between normal and targeted contrast-enhanced ultra sound parameters and MVD of transplanted tumors in nude mice at different stages was analyzed.Results: Targeted ultrasound contrast agent was successfully prepared;PI in 10 d,20d,30 d,40d MBt group was larger than that in MBc group,the diff erence was statistically significant(P < 0.05);TTP in 20 d and 30 d MBt group wa s smaller than that in MBc group,the difference was statistically significant(P <0.05);MVD in immunohistochemical counting was the highest in 30 d,and the diff erence was statistically significant among groups(P < 0.05);the overall correlation between PI and MVD in targeted ultrasound contrast group was systematic(P <0.05).The overall correlation between TTP and MVD was not significant(P > 0.05).The PI value of MBt group was positively correlated with MVD at 20 d and 30 d.The TTP value of MBt group was negatively correlated with MVD at 20 d and30 d(P < 0.05).Conclusion: MVD of ovarian cancer transplanted in nude mice is correlated with targeted contrast-enhanced ultrasound(CEUS)with CXCL12 antib ody,and the correlation is easily reflected in the early stage of transplanted ovaria n cancer(20 d,30 d).This indicates that the parameters of targeted contrast-enhan ced ultrasound can help to evaluate the growth of neovascularization in ovarian ca ncer to some extent.
