In this paper,fresh Momordica grosvenori was taken as the research object.First,the extraction of Momordica grosvenori was extracted with the extraction technology of normal temperature ultra high pressure(UHP).The optimum extraction process parameters were determined with the mogrosides V extraction rate,and compared with the traditional extraction method.At the same time,using the extraction rate and light transmittance of mogrosides V as the evaluation index,the clarifier was selected and optimized,and the best clarifier and clarification conditions were determined.Finally,through the exploration of the technological conditions for the preparation of the spherical silica gel column,the further purification of mogrosides V was completed and the high purity of the V was obtained.(1)The optimum extraction conditions of mogrosides were determined by ultra-high pres sure.Through single factor and orthogonal experiments,the best extraction parameters were determined treatment pressure 300 KPa,treatment time 2.5 min,liquid-solid ratio 1:12.Under this extraction condition,the mogrosides V extraction rate was 4.705 mg/g after repeated extraction for 3 times.The results also showed that the main factors affecting the extraction of mogrosides V were the liquid-solid ratio,and the second was the treatment pressure,and the effect of treatment time on the mogrosides V extraction rate was not significant.(2)The best clarifier and clarification conditions were determined.Through the roughing experiment,four kinds of clarifier with good clarification effect were screened out,and single factor and orthogonal optimization were carried out respectively.The optimum clarification process parameters of ZTC-Ⅱ clarifier are as follows:the dilution extract is 0.67 times the concentration of the original solution,and the proportion of A and B components(mL:mL)is 0.67:2.33 and the addition dosages is 2%.Under this clarification conditions,repeated clarification 3 times,the overall desirability is 98.21%.The optimum clarification process parameters of JH clarifier are as follows:the dilution extraction solution was 1.25 times the concentration of the original solution,and the proportion of A and B components(mL:mL)was 2.00:3.00 and the addition dosages was 4%.Under this clarification conditions,repeated clarification 3 times,the overall desirability is 96.65%.The optimum clarification process parameters of the 101 clarifier are as follows:the dilution extract is 1.25 times the concentration of the original liquid,the reaction temperature is 30℃,and the addition dosages is 1.5 g/100 mL.Under this clarification condition,repeated clarification 3 times,the overall desirability is 93.15%.The optimum clarification process parameters of Ca(OH)2 are as follows:the addition dosages is 1.5 g/100 mL,the reaction temperature is 30℃,stirring speed 20 r/min.Under this clarification conditions,repeated clarification 3 times,the overall desirability is 97.58%.By comparing the results of the clarification test of four kinds of clarifier,we can see that the clarifying effect of ZTC-Ⅱclarifier is the best and the process is stable.It can be used for clarification of the extracts of mogrosides.(3)The optimum technological conditions for purification of mogrosides V using spherical silica gel(250×30 mm)were determined.Using spherical silica gel to further purify 50%the extracts of mogrosides V,and the optimum process parameters were determined by the experiment:flow rate:15 mL/min,sample volume 1.0 mL,and elution conditions:23%acetonitrile-77%water(V:V).Under the conditions of preparation,the purity of mogrosides V reached 96.1%and the recovery rate was 95.0%by HPLC.High purity products were obtained. |
