| BackgroundSystemic lupus erythematosus(SLE)is a systemic autoimmune disease that violates skin and multiple-organs.Some unknown causes induce the body to produce a variety of autoantibodies,resulting in autoantibodies and the corresponding auto-antigen binding for the immune complex,through the type III hypersensitivity and damage their own tissues and organs.Most SLE patients were from20 to 40 years old,and more than 90% are women,which seriously affected the physical and mental health of women of child-bearing age.The etiology and pathogenesis of SLE has not yet clear.Recent studies suggest that omentin-1,adiponectin and resistin are associated with various autoimmune diseases such as SLE and rheumatoid arthritis(RA).At present,there are few studies on the association of the changes of omentin-1,adiponectin and resistin which SLE,as well as the omentin-1,adiponectin and resistin genes polymorphisms and SLE genetic susceptibility.ObjectiveTo evaluate the association of omentin-1,adiponectin and resistin gene single nucleotide polymorphisms(SNPs)with systemic lupus erythematosus(SLE)susceptibility.MethodsIn our study,four SNPs(rs2274907,rs35779394,rs79209815,rs13376023)of omentin-1,two SNPs(rs16861194,rs266729)of adiponectin and three SNPs(rs1862513,rs3745368,rs3745367)of resistin were genotyped in a cohort of 593 Chinese SLE patients and 643 controls.The genotyping was conducted using TaqMan SNP genotyping assays.ResultsNo significant differences were observed for the distribution of allele and genotype frequencies of the above SNPs between SLE patients and controls.The genotype effects of recessive,dominant models were analyzed,and we found an increased risk in rs3745368 polymorphism under the dominant model(GG vs.AA + GA,P = 0.026).In addition,we also found that the GG genotype frequencies of the rs13376023 in omentin-1 were associated with oral ulcers in SLE patients(P = 0.021).In resistin,the GG genotype and G allele frequencies of the rs3745367 polymorphism were significantly higher in SLE patients with renal disorder(P = 0.034,P = 0.016,respectively),and the GG genotype and G allele frequencies of the rs3745368 polymorphism were both associated with discoid rash in SLE patients(P = 0.037,P = 0.011,respectively).Haplotype analyses indicated that the haplotype CGA for resistin was significantly associated with SLE susceptibility(P = 0.005).ConclusionsOmentin-1,adiponectin and resistin SNPs are not associated with genetic susceptibility to SLE,but may contribute to some specific clinical phenotype of this disease.Further studies are necessary to explore the precise roles of omentin-1,adiponectin and resistin gene in the pathogenesis of SLE. |
