Objective:By measuring the level of CCL2 in peripheral blood of patients with chronic insomnia and mild cognitive impairment, the relationship between chronic insomnia and mild cognitive impairment and the level of CCL2 in peripheral blood was investigated. It provides a new research target for early intervention and early prevention of chronic insomnia with mild cognitive impairment. Methods:Selected patients with chronic insomnia in the Department of Geriatrics,General Practice, and Department of Neurology at the First Affiliated Hospital of Bengbu Medical College. All patients met the US Diagnostic and Statistical Manual for Mental Disorders Version 5 CID diagnostic criteria and applied the Montreal Cognitive Assessment Scale(Montreal).Cognitive Assessment(Mo CA)evaluated their overall cognitive function and grouped them, and collected normal controls matched with gender, age, and educational level from the physical examination center.Fasting venous blood was taken by enzyme-linked immunosorbent assay to detect serum CCL2 concentration.Results:1.There was no significant difference in gender composition, age and education years between the three groups(p>0.05).2.The comparison of Mo CA scores among the three groups was statistically significant(P<0.05).Comparison between groups showed that the scores of the chronic insomnia group and the cognitive impairment group were significantly lower than the control group, and the MCI group had a lower score than the CID group.3.There were significant differences in the peripheral blood CCL2 levels,PSQI scores, and HAD scores among the three groups(P<0.05).Comparison between groups showed that the levels of chronic insomnia and cognitive impairment groups were significantly higher than those of the control group, and the CCL2 level in the MCI group was higher than that in the CID group.4.The cognitive function of each group compared the factors,CID group and MCI group as the spatial executive function, delayed recall, attention score was lower than the control group,MCI group also had a language score lower than the CID group.(P<0.05)5.Peripheral blood CCL2 levels were positively correlated with HAD scores,PSQI scores, and negatively correlated with Mo CA scores, visual space executive function, delayed recall, attention, language ability, abstraction ability, and naming.(P<0.05)6.Multiple linear regression analysis was performed using CCL2 as the dependent variable and Mo CA factors as the independent variables. The results showed that the independent risk factors of CCL2 were visual space, delayed recall, attention and language, and were negatively correlated.(P<0.05)Statistically significant7.CCL2 was the dependent variable and multiple linear regression analysis was performed on the factors of age of education,PSQI,Mo CA,and HAD.The results showed that the independent risk factors of CCL2 were PSQI,Mo CA,and were positively correlated with PSQI and negatively correlated with Mo CA.(P<0.05)Statistically significant.。Conclusion:1.chronic insomnia can lead to decreased cognitive function, including the main performance of visual space, delayed memory, attention, and the deeper the insomnia, the worse the cognitive function.2.Patients with chronic insomnia and chronic insomnia associated with mild cognitive impairment have increased CCL2 levels in peripheral blood, suggesting that CCL2 may be involved in the process of chronic insomnia and provide new research directions for the study of chronic insomnia.3.The level of CCL2 in peripheral blood of patients with chronic insomnia and mild cognitive impairment is significantly increased.4.Under the premise of multivariate stepwise regression analysis, depending on spatial execution power, language function, attention, and delayed memory are risk factors for serum MCP-1,suggesting that CCL2 may be involved in the process of cognitive impairment in patients with chronic insomnia and be chronic Insomnia causes cognitive impairments to provide new research targets.1.Chronic insomnia can lead to decreased cognitive function. The main manifestations are visual space, delayed recall, and attention. The deeper the degree of insomnia, the worse the cognitive function.2.Patients with chronic insomnia and chronic insomnia with mild cognitive impairment have elevated CCL2 in peripheral blood, suggesting that CCL2 may be involved in the process of chronic insomnia, providing new research directions for the study of chronic insomnia.3.The level of CCL2 in peripheral blood of patients with chronic insomnia and mild cognitive impairment is significantly higher. After multiple stepwise regression analysis excludes confounding factors, visual astronomical performance, language function, attention and delayed memory are peripheral blood CCL2 hazards.Factors suggest that CCL2 may be involved in the process of cognitive impairment in patients with chronic insomnia, and provide a new research target for cognitive impairment caused by chronic insomnia.
