Objective:To investigate the changes and effects of Helios~+regulatory T cells in peripheral blood of patients with acute coronary syndrome(ACS).Methods:A total of 146 patients were recruited in this study,divided into acute coronary syndrome(ACS)group(n=61),stable angina pectoris(SA)group(n=30),coronary atherosclerosis(CA)group(n=30)and control group(n=25).To analyze the clinical data of all patients.The peripheral blood samples were collected and isolated by density gradient confriguration method.The frequency of Helios~+Treg among CD4~+T cells was analyzed using flow cytometry.Treg cells in peripheral blood were obtained by magnetic cell sorting(MACS),the expression of Helios mRNA in the purified Treg cells was determined by real time-PCR.The plasma levels of TGF-β1、IL-10、hs-CRP and IL-6 were determined by ELISA.Results:The frequency of Helios~+Treg in CD4~+T cells was significantly lower in ACS group compared to SA group,CA group and Control group(p<0.01).Correspondingly,the expression of Helios mRNA in ACS group was significantly lower than those in SA,CA and Control group(p<0.01).Compared with other three groups,the concentrations of hs-CRP and IL-6 were significantly higher in patients with ACS.The frequency of Helios~+Treg was negatively correlated with plasma levels of hs-CRP(r=-0.207,p=0.039),but had no correlation with IL-6 levels in plasma(r=-0.161,p=0.109).By contrast,the concentrations of TGF-β1 and IL-10were significantly lower in patients with ACS.The frequency of Helios~+Treg was positively correlated with plasma levels of TGF-β1 and IL-10(r=0.323,p=0.001/r=0.278,p=0.005).Conclusion:The proportion of Helios~+Treg cells in peripheral blood of patients with acute coronary syndrome was significantly reduced,the balance between pro-inflammatory and anti-inflammatory factors was broken,and the immunosuppressive effect of Helios~+Treg decreased,resulting in progression of the disease.
