| Objective:To observe the expression of CTRP9(c1q-tumor necrosis factor-related protein-9)in the rat model of right ventricular outflow tract obstruction(RVOTO),and to investigate the correlation between right ventricular hypertrophy fibrosis and the expression of CTRP9.Methods: Thirty male SD rats were randomly divided to three groups: right ventricular outflow tract obstruction(RVOTO)group,sham group and control group.There were 10 rats in each group.The RVOTO group were performed pulmonary arterial constriction(PAC).The sham group were performed thoracotomy without PAC.The control group were not accepted any surgical operation.The heart specimens were harvested after 4 weeks.The expression of CTRP9 in plasma was detected by Enzyme-linked immunosorbent assay(ELISA),and the thickness of the right ventricular myocardium was measured by Thickness-Gauge..The volume ratio of collagen fibers in the right ventricular myocardium was calculated by the software of image pro plus(IPP)which dyed by MASSON.The expression of CTRP9 in myocardial tissue section was detected by immunohistochemical method.The concentration of CTRP9 in the right ventricular myocardium was detected by western-blot.T test and person correlation analysis and linear fitting were used to analyze the experimental results.Results:1.Right ventricular outflow tract obstruction model: average thickness of right ventricular in operation group,sham group and control group show that operation group thicker than other groups(operation group:(0.938 + 0.938)mm;sham group:(0.260 + 0.260)mm;control group:(0.249+0.249)mm,p=0.000).we partitioned the Right ventricular myocardium into night areas,and measured each area.The result show that the hypertrophy of the right ventricle was most thickened with outflow tract.2.The rate of myocardial fibrosis in right ventricle:Myocardial collagen fiber volume ratio showed that the right ventricular hypertrophy and the collagen fiber volume ratio in RVOTO rats were obvious higher than sham group and control group(p=0.000).There was no statistically significant difference between the sham operation group and the control group.Morphology was characterized by hypertrophy of myocardial cells and disorder of collagenous fibers.3.Concentration of CTRP9 in plasma: The concentration of CTRP9 in plasma showed no statistically significant difference between the sham operation group and the control group(p=0.956).The concentration of CTRP9 in the operation group was statistically significant compared with the sham operation group and the control group(p=0.000). Pearson correlation analysis showed that the correlation coefficient was-0.910,indicating that the concentration of plasma CTRP9 was highly correlated with the fibrosis rate.4.The concentration of CTRP9 in myocardial tissue: Immunohistochemical method showed that the concentration of CTRP9 in the operation group was significantly higherthan that in the sham operation group and the control group,and the difference wasstatistically significant(p=0.000).The concentration of CTRP9 in myocardial tissue was lso indicated,and the concentration of CTRP9 in myocardial tissues of the operation roup was significantly increased,and the difference was statistically significant(p =0.000) ompared with the sham operation group and the control group.Pearson correlation was sed to analyze the relationship between the fibrosis rate and the concentration of CTRP9, nd the correlation coefficient was 0.947 and 0.950,indicating that the degree of yocardial fibrosis was highly correlated with the concentration of the control group.· Conclusion: In the right ventricular outflow tract obstruction animal model,the xpression of CTRP9 was significantly reduced in the circulation system.However,the xpression in myocardial tissue increased.It is indicated that in the right ventricular econstruction process,CTRP9 may actively participate in the pathological changes of the ight ventricular hypertrophy. |
