Breast cancer is one of the most common malignancies in women.At present,the treatment of breast cancer includes surgery,chemotherapy,radiation therapy,molecular targeted therapy.But there are still some problems in these ways.We have to find a new method or medicine to solve the problem.Vitamin K2 is a fat-soluble Vitamin naturally.Its main function is to treat osteoporosis and anemia.Recent studies have found that Vitamin K2 can suppress tumor cell growth.However,the effect of Vitamin K2 on breast cancer is still unknown.Therefore,this paper pay attention to the role of Vitamin K2 on human breast cancer cell line and molecular mechanisms.The cell vitality of MCF-7 was remarkably decreased after the prolonged treatment with 100 μM Vitamin K2.We adopt different concentrations of Vitamin K2 treatment for 24 hours,the cell vitality also decreased.The results indicated that Vitamin K2 exhibited inhibitory growth effect on MCF-7.It was found that Vitamin K2 induced MCF-7 cell apoptosis by flow cytometry experiment.The number of apoptotic cells in the late stage was significantly higher than that in the control group..In order to further explore specific mechanism of the apoptosis of breast cancer,we studied the total ROS,mitochondrial ROS,mitochondrial membrane potential.It was found that Vitamin K2 induced to produce a large number of ROS,which stem from mitochondrial and caused mitochondrial membrane potential decreased.By adding ROS inhibitor NAC,the cell viability was significantly enhanced.Therefore,Vitamin K2 induced MCF-7 cell apoptosis through mitochondrial pathway.Finally,Vitamin K2 didn’t affect the vitality of normal cells and its ROS level.In conclusion,VitaminK2 can induce apoptosis of cancer cells.But it does not affect the normal cell vitality.So it may become a new type of small molecule drug for breast cancer. |
