gastric carcinoma.Methods:The cancer tissues and adjacent normal tissues of 27 patients with gastric cancer undergoing gastric cancer resection in the Second Hospital of Hebei Medical University from March 2017 to October 2017 were selected.First,HE staining was used to determine the degree of differentiation and types.Then immunohistochemistry and Western blot were used to analyse the expression of FOXO1,Bim and P27 in the cancer tissues and the normal tissues adjacent to the cancer.The relationships between the expression of proteins and the clinical characteristics of gastric cancer were discussed.Results:1.Group of patientsThe final 27 patients were included in the analysis,including 17 male patients and 10 female patients,age: 30~77 years old,average age:(58.92±8.62)years old.2.Results of HE stainingHE staining showed that the pathological type of gastric cancer patients was mainly adenocarcinoma.The cells in the normal tissue adjacent to the carcinoma were arranged in neat arrangement.3.Results of IHCThe positive expression rates of FOXO1 in gastric cancer tissues and normal tissues adjacent to cancer were 74.07%(20/27)and 22.22%(6/27),respectively.The expression of gastric cancer tissues was higher than that of normal tissues adjacent to the cancer,and the difference between the two was statistically significant(P<0.05).In gastric cancer patients,the degree of differentiation was: the positive expression rate of low differentiation was 55.56%(5/9),the positive expression rate of medium differentiation was 70%(7/10),the positive expression rate of high differentiation was 100.00%(8/8),and the difference between the three groups was statistically significant(P<0.05).The positive expression rates of Bim in gastric cancer tissues and normal tissues were 29.63%(8/27)and 85.19%(23/27),respectively.The positive expression rate of gastric cancer tissues was lower than that of normal tissues adjacent to the cancer,and the difference between the two was statistically significant(P<0.05).In the patients with gastric cancer,the positive rate of lymph node metastasis was 16.67%(3/18),and the positive rate of gastric cancer without lymph node metastasis was 55.56%(5/9).The difference between the two groups was significant,and the difference was statistically significant(P<0.05).The positive expression rates of P27 in gastric cancer tissues and normal tissues were 40.74%(11/27)and 81.48%(22/27),respectively.The expression of gastric cancer tissue was lower than that of normal tissue adjacent to the cancer,and the difference between the two was statistically significant(P<0.05).In the patients with gastric cancer,the positive rate of lymph node metastasis was 22.22%(4/18),and the positive rate of gastric cancer without lymph node metastasis was 77.78%(7/9).The positive rate of the stage I+II was 16.67%(2/12),and the positive expression rate in stage III+IV was 60%(9/15).The positive rate of no lymph node metastasis group and pathological stage III+IV group were higher than that of the control groups,and the differences were statistically significant(P<0.05).4.Results of Western blotThe protein expression level of FOXO1 in gastric cancer cells and normal tissues adjacent to cancer were(0.33±0.04)and(0.22±0.03),respectively.The expression of gastric cancer tissue was higher than that of normal tissue adjacent to the cancer,and the difference between the two was statistically significant(P<0.05).In gastric cancer patients,the degree of differentiation was:the protein expression level of low differentiation was(0.30±0.01);the protein expression level of medium differentiation was(0.34±0.03);the protein expression level of high differentiation was(0.36±003),and the difference between the three groups was statistically significant(P<0.05).The protein expression level of Bim in gastric cancer tissues and normal tissues were(0.31±0.07)and(0.71±0.06),respectively.The protein expression level of gastric cancer tissue was lower than that of normal tissue adjacent to the cancer,and the difference between the two was statistically significant(P<0.05).In the patients with gastric cancer,the protein expression of lymph node metastasis was(0.29±0.07);and the protein expression of gastric cancer without lymph node metastasis was(0.35±0.03).The difference between the two groups was significant,and the difference was statistically significant(P<0.05).The protein expression level of P27 in gastric cancer tissues and normal tissues were(0.48±0.08)and(0.71±0.07),respectively.The expression of gastric cancer tissue was lower than that of normal tissue adjacent to the cancer,and the difference between the two was statistically significant(P<0.05).In the patients with gastric cancer,the protein expression of lymph node metastasis was(0.44±0.08),and the protein expression of gastric cancer without lymph node metastasis was(0.55±0.05).The protein expression of the stage I+II was(0.42±0.07);and the protein expression level in stage III+IV was(0.53±0.06).The protein expression of no lymph node metastasis group and pathological stage III+IV group was higher than that of the control group,and the difference was statistically significant(P<0.05).Conclusion:1.The expression of FOXO1 protein in the cancerous tissues was higher than that of the normal tissues adjacent to the cancer.The expression of FOXO1 in gastric carcinoma is related to the differentiation degree and the higher the degree of tumor differentiation,the higher the expression of FOXO1 was.It is suggested that FOXO1 may play a role in the carcinogenesis of normal gastric mucosa,and it can be used as a target of target therapy for gastric cancer.2.The expression of Bim protein was lower than that of normal tissues adjacent to the carcinoma.It is suggested that Bim may inhibits the development of gastric cancer as a tumor suppressor gene.The expression of Bim in gastric carcinoma is related to lymph node metastasis,and no lymph node metastasis group was higher than that of lymph node metastasis group.3.The expression of P27 protein was lower than that of normal tissues adjacent to the carcinoma.It is suggested that P27 may inhibits the progress of gastric cancer.The expression of P27 in gastric carcinoma is related to lymph node metastasis and TNM staging.No lymph node metastasis group,TNM stage III +IV group were higher than those in the corresponding control groups.4.FOXO1 inhibits the development of gastric cancer in cell apoptosis and cell cycle through the action of Bim
