In cum uncis Rubiales belonging to the Rubiaceae genus Uncaria rhynchophylla stem hook;It is widely distributed in China and has high medicinal value;It has many pharmacological activities,such as anti-inflammatory,analgesic,anti tumor,anti platelet aggregation and so on.It is also used as a kind of food and health care products in the folk.The active component is unstable,and it will be destroyed when it is continuously heated to change the content.In this paper,based on the drying and traditional decocting of Uncaria Rd.,the content change of its effective components was studied,and the relationship between the anticoagulation and the effect of drying process was preliminarily constructed.In this paper,10 drying models were used to simulate drying curves of Chinese medicinal materials Uncaria at 45、55、65 and 75℃;The correlation coefficient(R2),chi square(X2)and root mean square error(RMSE)were used to evaluate the goodness of fit.Finally,the best drying model of Uncaria was selected by comprehensive analysis of the model constants,at the same time,the kinetic parameters are measured.The results of the study show that:The dry model Newton is the best drying model for Chinese medicinal herbs.The range of D value of water diffusion coefficient is from 1.96×10-10 to 5.18×10-10 m2 s-,the activation energy in the drying process is 30.37kJ/mol.Obtaining the chromatograms of different time points in the drying and decocting process by HPLC,the peak area of 4 characteristic peaks(1.rhynchophylline,2.Unknown compound,3.isorhynchophylline,4.ISO runcarine)was used to study the change rule.The experimental results show that In the two processes,the peak area of the 4 characteristic components increased first and then decreased,and the maximum value of the 30min and the constant temperature drying570min reached the maximum.At the same time,the increase segment of its content is linearly fitted by the statistical software,the experimental results show that the 4kinds of characteristic components have a good linear relationship with the time in the increase of the content of the content.Based on the rising segment of the characteristic component content in the drying experiment,the anticoagulant anticoagulant study was carried out at 7 time points in vitro to obtain the blood sample coagulation time,then the relationship between the peak area of the 4 characteristic components in the HPLC chromatogram of dry samples and the reference to the blood coagulation time of the corresponding samples was investigated and the quantity effect relationship was constructed;At the same time,the relationship between the HPLC chromatogram and the blood coagulation time was investigated with the peak area of 4 characteristic components,and the preliminary construction of the spectral effect relationship of the anticoagulant anticoagulant in vitro was preliminarily constructed.The results showed that there was a good linear relationship between the coagulation time and the peak area of the sample in the rising stage of drying process(the correlation coefficient was greater than 0.9);By analyzing the equation of spectral efficiency relationship,we can know that 4 characteristic components,rhynchophylline and isorhynchophylline,are the main active substances of Chinese medicine Uncaria.
