Objective:To verify the residual tumor cells in the marginal sub-hypothermia area lead to recurrence after argon helium cryoablation of the hepatocellular carcinoma,and explore the feasibility of improving the therapeutic effect of argon helium cryoablation by combining with arsenic trioxide.Methods:Complete the argon helium cryoablation of H22 cells cultured in the ascites of the Kunming mice in vitro with-20oC,-40oC,-60oC respectively,and keep the one group of cells in 37oC.Observe the microstructure of the disposed cells by transmission electron microscope.Take other four groups of Kunming mice to get subcutaneous injection with the disposed cells respectively.Divide each group into the Normal Saline(NS)subgroup that receive the NS treatment and the Arsenic trioxide(As2O3)subgroup receive the arsenic trioxide chemotherapy for one week.Test the blood ALT level and lymphocyte sub-population,and observe the growth of the tumors.Kill the mice,dissect the tumor and complete the pathological examination.Result:1.Argon helium cryoablation causes microstructural changes in tumor cells,which is related to the temperature.2.The disposed cells in each group develop into tumors in mice,which is inhibited by cryoablation and arsenic trioxide.The inhibition is correlated to ablation temperature.3.Both cold ablation and arsenic trioxide increase the percentage of CD4+T cells and the proportion of CD4+/CD8+,which is correlated to ablation temperature.Conclusion:1.The temperature in the marginal sub-hypothermia area of tumor during the hepatocellular carcinoma cryoablation therapy was not low enough,the residual tumor cells in which area cause tumor recurrence after cryoablation.2.The argon helium cryoablation could play a synergistic role in the treatment of hepatocellular carcinoma with arsenic trioxide,the regulation of anti-tumor immunity might to be one of the mechanisms of which.
