Objective: To purify and identify the secondary metabolites of two actinomycetes,and to analyze the bioactive components of the two actinomycetes.Methods:(1)By using the methods like silica gel column chromatography,Sephadex gel molecular sieve chromatography and HPLC preparation,secondary metabolites were isolated and purified;(2)High resolution mass spectrometry and nuclear magnetic techniques were used to identify the compounds.Combining the UV absorption peaks,their structures could be speculated or determined.The secondary metabolites of the Streptomyces sp.FJS31-2 were detected by mass spectrometry and the secondary metabolites were systematically analyzed;(3)The antibacterial activity of the compound was determined and quantified by filter paper diffusion method and double dilution method.Results:(1)From the secondary metabolites of Streptomyces sp.FJS31-2,eight compounds were isolated and their relative molecular mass vary from 1200 to 1400.They were predicted to be new II type polykethers.The secondary metabolites in 22 samples were predicted;(2)From the secondary metabolites of Actinomadura sp.FXJ1.344,two compounds were isolated and identified as 6-Hydroxytetrangulol and tetrangulol.Antibacterial activity experiment showed that antibacterial circles of the two compounds(50 μg/m L)on Micrococcus luteus were 16 mm and 14 mm;the minimum inhibitory concentrations(MIC)of the two compounds were 3.90 μg/m L and 15.6 μg/m L respectively.Conclusion:(1)Streptomyces sp.FJS31-2 produced a variety of polyketides through secondary metabolism.Complicated secondary metabolism spectrum showed that it had great development value;(2)Actinomadura sp.FXJ1.344 could produce Angucyclinone compounds;the bacteriostatic activities of two specific compounds were first discovered,their antibacterial activity on Micrococcus luteus indicated that they could have certain development value as lead compounds in drug discovery;and the experiments proved that biological fermentation production of tetrangulol and 6-hydroxytetrangulol may have certain feasibility.
