Objective:To investigate the results of inspection reports in our hospital in children with acute diarrhea pathogenic microorganism,analysis of the composition of pathogenic microorganisms,to provide theoretical reference for the prevention and treatment of the disease.By viral culture,identification,concentration and purification,establish a stable and uniform rotavirus strain library.Using the virus library build human rotavirus animal model of rats with gastric attack poison rats and mice to observe the diarrhea,tissue pathological changes and a series of changes,to explore the pathogenic mechanism of rotavirus.Methods:2014 year in our hospital 422 cases of acute diarrhea disease in children patients as the research object,collecting stool samples of children,bacterial culture and identification,detection and evaluated by ELISA virus antigen,reverse transcription polymerase chain reaction for detection of viral nucleic acid of virus,type,distribution characteristics and the related statistical properties of pathogenic microorganisms.Selected specimens of rotavirus diarrhea purification develop rotavirus,rotavirus against poison library is established.Only 3 days of age 50kunming rats by rotavirus vaccination or not were randomly divided into experimental group(40)and the control group(40),the experimental group by gastric perfusion rotavirus(TJTG-986)culture,the control perfusion culture contains no virus,respectively by gastric observe different time points after inoculation diarrhea,stool rotavirus antigen ELISA method for detection,and take its then we observed the changes of intestinal tissue morphological changes and rotavirus engraftment,electronic microscope cell ultrastructural changes.Results:the incidence rate of different ages of children have significant difference(χ~2=158.083,P<0.05),less than 2 years of age infants easy onset detection in children with acute diarrhea;pathogenic microorganism in 254 cases,the positive rate was 60.19%,the detection of virus in 197 cases(77.56%),rotavirus detection in146 cases(57.48%),48 cases of pathogenic bacteria(18.90%),9 cases of fungi and parasites(3.54%);the gender had no significant effect on the incidence of acute diarrhea in children,but the impact of pathogenic microorganism infection by season.Conclusion:acute diarrhea pathogenic microorganism in children is mainly by virus,rotavirus is most,multiple in the winter,to understand the distribution of pathogenic microorganisms has an important guiding significance for the prevention and treatment of the disease.pathogenic microorganism in 254 cases,the positive rate was 60.19%,the detection of virus in 197 cases(77.56%),rotavirus detection in 146cases(57.48%),48 cases of pathogenic bacteria(18.90%),9 cases of fungi and parasites(3.54%);the gender had no significant effect on the incidence of acute diarrhea in children,but the impact of pathogenic microorganism infection by season.Through the separation and purification of clinical specimens of viral diarrhea,the establishment of a regional human rotavirus strains.Rotavirus rats model is successfully established,the experimental group was observed in all 24 h after inoculation of virus diarrhea,4 d after inoculation of peak and feces ELISA detection of RV antigen are all positive,the control group were not found diarrhea,RV antigen test negative.Highlight the pathological changes of intestinal epithelial cells widely vacuoles degeneration.Under the electron microscope villi epithelial cell damage,loose connections,broadening,integrity is damaged.TJTG-986 strain library build successful,subculture is stable and can be successfully infected newborn kunming rats model of rotavirus infection.Rotavirus diarrhea may be because of rotavirus in fixed value growth on small intestinal epithelial cells,causing intestinal epithelial cell vacuoles degeneration necrosis,intestinal epithelial cell death brings a series of pathological changes,cause indigestion,causes watery diarrhea.
