Cancer has become one of the leading causes of death in the world’s population,and the incidence and mortality of lung cancer are at the top of the list and are on the rise year by year.The development of safe and effective anti-tumor drugs has become a research hotspot in the world medical medicine field.The purpose of this thesis is to systematically study the extraction and effect screening of Camellia nitidissima extract,as well as its preventive and therapeutic effects on urethane-induced lung cancer in mice,and to further explore its anti-tumor mechanism in vitro,and finally to golden tea.The safety of flower extracts was evaluated.The specific research content and results are as follows:(1)The ultrasonic-assisted ethanol extraction method was used to prepare the Camellia nitidissima flower extract.The results showed that the extraction rate of50 % ethanol concentration was the highest,reaching 18.62 %,and the inhibitory effect of the concentration of ethanol extract of Camellia oleifera extract on A549 lung cancer cells Preferably,the inhibition rate of A549 lung cancer cells is as high as50 % at a dose of 40 μg/mL.(2)The animal lung cancer model experimental results found that the incidence of lung cancer in the male and female mice of the urethane model group was 93.57 %and 89.47 % respectively.The Camellia nitidissima extract can reduce the incidence of lung tumor in the male and female mice to 80.00 %,respectively.77.78 %,and significantly increased the content of CAT,SOD,IL-2,TNF-α in mice of the model group and reduced the level of MDA,which proves that it can increase the body’s anti-oxidation level and enhance the body’s immune system function.(3)PI/Hoechst 33342/DNA fragmentation electrophoresis,and Annexin V-FITC/PI double staining experiments fully proved that the extract of Camellia nitidissima had the effect of inducing apoptosis in A549 cells.In addition,A549 cells treated with Camellia can effectively block the cell cycle of A549 cells.The results of detection of MMP,Caspase-3/8/9 and Bcl-2/Bax protein expression showed that the way of inducing A549 cell apoptosis was mitochondrial apoptosis pathway.(4)The in vitro CCK-8 experiment results showed that the Camellia nitidissima extract had no cytotoxicity to human normal liver cell.In vivo subchronic toxicity test results showed that 100,200,and 400 mg/kg of Camellia nitidissima extract had no significant effect on body weight,organ coefficient(heart,liver,spleen,lung,kidney),liver,kidney and spleen morphology of mice.In summary,this study preliminary explored the mechanism of the anti-tumor effect of Camellia nitidissima extract,and conducted a preliminary safety evaluation of the Camellia nitidissima extract,which provided a theoretical basis for the prevention and treatment of cancer in Camellia nitidissima. |
