The Expression Of Kiss-1 And Kiss-1R In Gastric Cancer And Its Clinical Significance | | Posted on:2019-01-16 | Degree:Master | Type:Thesis | | Country:China | Candidate:M Yang | Full Text:PDF | | GTID:2404330566479385 | Subject:Internal Medicine | | Abstract/Summary: | PDF Full Text Request | | Objective: Gastric cancer is a common digestive malignant tumor,which badly endangers people’s health.At present,the role of tumor invasion and metastasis in pathogenesis of gastric cancer has become a research hotspot.Kiss-1 is a tumor metastasis suppressor gene,and Kiss-1R is its receptor.There are many studies on the relationship between Kiss-1 and some tumors,but the role of Kiss-1 and Kiss-1R in the pathogenesis of gastric cancer is not clear.Therefore,this study detected the expression of Kiss-1 and Kiss-1R in gastric cancer and adjacent normal tissues,analyzed the correlation between the expression and clinicopathological data,and also analyzed the correlation between the two biological behaviors in gastric cancer.At the same time,the expression of Kiss-1 in AGS and SGC-7901 cell lines was detected,and its role in the pathogenesis of gastric cancer was analyzed.AGS cell line derived from gastric primary site of poorly differentiated adenocarcinoma,and SGC-7901 cell line derived from lymph node of metastatic gastric adenocarcinoma.Methods:Immunohistochemistry was used to detect the expression of Kiss-1and Kiss-1R in 40 cases of gastric cancer and 28 cases of adjacent normal tissues;western blot was used to detect the expression of Kiss-1in AGS and SGC-7901 cell lines.χ2 test was used for comparison of two groups.Correlation analysis was performed with spearman correlation analysis.All statistical analyses were performed using the SPSS 21.0software and P<0.05 was considered statistically significant.Results:1.The positive expression rate of Kiss-1 was statistically significant between gastric cancer group and adjacent normal group(P<0.01).The expression of Kiss-1 protein was significantly higher in gastric cancer with moderate and well differentiation compared to poor differentiation;significantly higher in gastric cancer without lymph node metastasis compared to lymph node metastasis;significantly higher in the low stage of gastric cancer(I + II)compared to advanced stage(III + IV).2.The positive expression rate of Kiss-1R protein was statistically significant between gastric cancer group and adjacent normal group(P<0.01).The expression of Kiss-1R in gastric cancer had no relation to clinicopathological data(P>0.05).3.Spearman rank correlation showed that the expression of Kiss-1in gastric cancer group was significantly and positively associated with the expression of Kiss-1R(r =0.312,P<0.05).4.The expression of Kiss-1 in metastatic gastric cancer cell line SGC-7901 was lower than that in primary gastric cancer cell line AGS.Conclusions:1.The expression of Kiss-1 protein in gastric cancer group was significantly down-regulated,which indicated that Kiss-1 playing a role of tumor suppressor in the occurrence of gastric cancer.2.The expression of Kiss-1 protein in gastric cancer group was down-regulated in group with low differentiation,lymph node metastasis,clinical stage III + IV,which indicated that tumor suppressor role of Kiss-1 in gastric cancer with higher malignant degree,stronger invasive and metastasis ability,poorer prognosis was obviously restrained.3.The expression of Kiss-1R protein in gastric cancer group was significantly down-regulated,and the expression of Kiss-1 was significantly and positively associated with the expression of Kiss-1R,which indicated that Kiss-1 was combined with Kiss-1R,involved in the pathogenesis of gastric cancer with synergistic effects.4.The expression of Kiss-1 in metastatic gastric cancer cell line was lower than that of primary gastric cancer cell line,suggesting that Kiss-1 was obviously restrained in the type of gastric cancer cell lines with high invasive ability. | | Keywords/Search Tags: | Gastric cancer, Kiss-1, Kiss-1R, Immunohistochemistry, Western-blot, Cell lines | PDF Full Text Request | Related items |
