With the development of the national economy and changes of modern life style,the incidence of hypertension and hyperlipidemia increased dramatically,these two kinds of chronic diseases seriously threaten the health of human beings.Compound Prunella vulgaris L.has been used for hypertension as a exact effect of agreement nosocomial prescription for many years,its effect of lowering blood pressure is good,and it has the function of diuresis and lowering blood lipid.In this study,we observed the influence of Extracts from Compound Prunella vulgaris L.on spontaneously hypertensive rats(SHR),normal SD rats and hyperlipidemia rats,investigated its antihypertensive,diuretic and hypolipidemic effects,and provided experimental basis for the clinical scientific application.This paper used the percolation method to extract the Compound Prunella vulgaris L.,the extraction technology was optimized by orthogonal experiment.The influence of Extracts from Compound Prunella vulgaris L.on the systolic blood pressure(SBP),diastolic blood pressure(DBP),heart rate(HR)and weight of SHR was studied by the tail cuff method during the eight-week continuous administration.Blood was taken from abdominal aorta of rats after administration of eight weeks.Then the serum angiotensinII(AngII),nitric oxide(NO),endothelin-1(ET-1)and atrial natriuretic peptide(ANP)content were measured respectively.After the rats were killed and the kidneys were removed,we weighed the weight of kidneys and calculated the renal dirty body ratio.The pathologic morphology changes of kidneys were monitored by using the optical microscopes.The effect of Extracts from Compound Prunella vulgaris L.on protein expression of the AngII,angiotensinII type1 receptor(AT1R)and connective tissue growth factor(CTGF)of SHR kidneys were analyzed by semi-quantitative immunohistochemical method.The influence of Extracts from Compound Prunella vulgaris L.on diuretic effect of normal SD rats was studied by Rat metabolic cage method after single administration of Extracts from Compound Prunella vulgaris L.;the influence was also studied of every day during seven-day continuous administration by the same method.At the same time,the effect of Extracts from Compound Prunella vulgaris L.on rat urine Na~+,K~+,and Cl~-concentration was also studied.The hyperlipidemia rat model was established by feeding high-fatty feeds.The influence of Extracts from Compound Prunella vulgaris L.on the hyperlipidemia rat was studied by measuring the levals of rat food intake,body weight,liver weight,liver coefficient,the content of the serum levels of total cholesterol(TC),glycerin three greases(TG),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C),calculating LDL-C/HDL-C and TG/HDL-C ratio,arterial stiffness index(AI).The experimental results showed that the optimum extraction process of Compound Prunella vulgaris L.:ethanol concentration 50%,soaking for 12 hours,flow rate was 2mL/min and 12 times the amount of solvent were percolation extraction.Extracts from Compound Prunella vulgaris L.could significantly lower the blood pressure(SBP and DBP)of SHR(P<0.05 or P<0.01),and dosage-effect relationship were observed during eight weeks.There was no significant effect on HR and body weight of SHR rats(P>0.05).The content of AngII and ET-1 were decreased,and the concentration of NO and ANP were increased in SHR’s serum(P<0.05 or P<0.01).There was no significant effect on the kidney weight and the renal dirty body ratio of SHR(P>0.05).The protein expression of AngII,AT1R and CTGF in kidney of SHR were significantly inhibited(P<0.05 or P<0.01).Extracts from Compound Prunella vulgaris L.had certain protective effect on kidney injury caused by elevated blood pressure.Extracts from Compound Prunella vulgaris L.could significantly increase the urine volume of normal SD rats(P<0.05 or P<0.01)and showed a significant dose-effect relationship.The concentration of sodium in the urine of rats was significantly increased(P<0.05 or P<0.01),while there was no significant effect on potassium and chlorine concentration(P>0.05),the diuresis mechanism may similar to weak acting diuretic.Extracts from Compound Prunella vulgaris L.had no significant effect on the food intake and body weight of hyperlipidemia rats(P>0.05).The levels of TC、TG、LDL-C、LDL-C/HDL-C、TG/HDL-C ratio and arterial stiffness index were significantly reduced(P<0.05 or P<0.01),and the levels of HDL-C were increased in serum of hyperlipidemia rats(P<0.05).Reduced liver weight and liver coefficient in hyperlipidemia rats(P<0.05or P<0.01).Extracts from Compound Prunella vulgaris L.shows obvious effects in antihypertension,diuretic and antihyperlipidemia.It has certain protective effect on kidney injury in SHR.It can decrease liver weight and liver coefficient in hyperlipidemia rats.Compound Prunella vulgaris L.may be more suitable for the clinical treatment of hypertension complicated with hyperlipidemia. |
