| [Objective] Epigenetic regulation plays an important role in the mechanism of tumor formation,including DNA methylation,histone covalent modification,chromatin remodeling,gene silencing and RNA editing.DNA methylation and demethylation formation the specific DNA methylation profiles.the methylation profile changes may lead to disease or tumors.Methyl-CpG binding domain(MBD)proteins is the "methylation reader",Methylation binding protein 2(MBD2)as one of the methyl CpG binding protein family members,because its knockout mice did not embryonic lethal,only affect lactation behavior in adulthood,it is considered a good target for the research and the application prospect.Previous studies in mice have shown that MBD2 knockdown affects the differentiation of T cells and delays the onset of acute lymphoblastic leukemia(T).But the detection of MBD2 among patients with hematological malignancies and the drugs that can regulate MBD2 expression are little seen,so we were collected and detected hematological malignancies and study the effect of Matrine on MBD2.[Methods] 1.The bone marrow samples from 200 initial diagnosed patients with hematological malignancies were collected,and the expression level of Methylation binding protein 2(MBD2)at mRNA level was detected by Real-time PCR method.2.Objective to investigate the effect of Matrine on the proliferation and apoptosis of B cell line Duadi,and to detect the expression of CpG binding protein 2(MBD2)at the level of mRNA after matrine treatment with Real-time PCR.[Results] 1.The expression of MBD2 in acute lymphoblastic leukemia was increased,which was statistically significant.Among them,B-ALL and poor prognosis group were significantly increased.The expression of MBD2 in female patients was increased,and the lower level of lactate dehydrogenase showed higher expression level of MBD2.Poor prognosis molecular biology showed higher MBD2 levels,OS,DFS showed no significant difference.2.In acute lymphoblastic leukemia,the expression of MBD2 was up-regulated and the activity of WNT pathway was up-regulated.3.Matrine can inhibit the proliferation and induce apoptosis of B lymphoma Duadi,and can down regulate the expression of MBD2.[Conclusions] 1.The expression of MBD2 was increased in B-ALL and poor prognosis group,and the expression of MBD2 was higher in female patients and patients with low lactate dehydrogenase.OS,DFS did not find significant differences.It may be a potential indicator of poor prognosis in ALL.Upregulation of MBD2 and WNT pathways in B-ALL may be a potential target for ALL therapy.2.Matrine can inhibit the proliferation and induce apoptosis of Duadi,and can down regulate the expression of MBD2. |
