| Cyclocarya paliurus(Batal)Ijinskaja,which is a unique species of Chinese plant,also known as Yaoqianli,copper qian tree.The ancient book records its branches and leaves have the effect of clearing heat and detoxifying,and it is usually made into tea by the people.At present,a large number of studies have shown that Cyclocarya paliurus(Batal)Ijinskaja has the functions of reducing blood glucose,reducing blood lipid and antioxidant.Zymoyic fungal powder of cordycepes sinensis is obtained by separating the natural cordyceps sinensis strain into deep fermentation.It has been reported that the zymoyic fungal powder of cordycepes sinensis has the same function as cordyceps sinensis,such as improving immunity and anti-tumor.In this paper,selenium-rich cyclocarya paliurus and zymoyic fungal powder of cordycepes sinensis was used as main ingredients to made compound preparations,using diabetic mice to evaluate its hypoglycemia,antioxidation and immunomodulatory efficacy.And the antioxidant activity by the reaction system of diphenyl bitter acyl radical(DPPH),hydroxyl radical(·OH)and superoxide anion(O2·-)in vitro were evaluated.Finally,the effect of the compound preparationson the survival rate of the insulinoma,damaged by streptomycin was studied.The main research works are as follows.(1)The selenium-rich Cyclocarya paliurus(Batal)Ijinskaja compound was obtained by using hot water and ethyl alcohol to extract selenium-rich Cyclocarya paliurus(Batal)Ijinskaja.Selenium-rich Cyclocarya paliurus(Batal)Ijinskaja compound extract,zymoyic fungal powder of cordycepes sinensis,wolfberry extract and Poria cocos extract are made into tablet,oral liquid and beverage.The selenium content of levers of the compound preparations was measured by hydride generation-atomic fluorescence spectrometry.The adenosine content of levers of the compound preparations was measured by HPLC.The results showed that the selenium content of the compound preparations was79.4μg/g and the adenosine content of the compound preparations was 895μg/g.(2)According to the recommended dose,different doses of the compound preparations selenium-rich cyclocarya paliurus and zymoyic fungal powder of cordycepes sinensis were gave to the diabetic mice induced by giving high fat diet and the intraperitoneal injection of streptozotocin.After 42 d of administration,The levels of oral glucose tolerance,serum antioxidant enzyme activities and immune activities of diabetic mice were measured.The results showed that the appropriate dose of selenium-rich cyclocarya paliurus and zymoyic fungal powder of cordycepes sinensis compound preparations could significantly reduce postprandial blood glucose,increase GSH-Px activity and the spleen index.(3)The antioxidant activity of selenium-rich cyclocarya paliurus and zymoyic fungal powder of cordycepes sinensis compound preparations was measured by the reaction system of diphenyl bitter acyl radical(DPPH),hydroxyl radical(·OH)and superoxide anion(O2·-).The results showed that selenium-rich cyclocarya paliurus and zymoyic fungal powder of cordycepes sinensis compound preparations could effectively eliminate free radicals.(4)The effect of selenium-rich cyclocarya paliurus and zymoyic fungal powder of cordycepes sinensis compound preparations on the survival rate of the NIT-1 insulinomaβcell damaged by streptozotocin was measured.ROS level of the NIT-1 insulinomaβcell was measured.The results showed that the appropriate dose of selenium-rich cyclocarya paliurus and zymoyic fungal powder of cordycepes sinensis compound preparations could improve the NIT-1 insulinomaβcell survival rate,and significantly lower intracellular ROS levels.At present,most of the selenium-rich cyclocarya paliurus products developed in the market are drinks.No literature has been reported about selenium-rich cyclocarya paliurus with other traditional Chinese medicine The results of this paper might provide important experiment evidence for the research and development in selenium-rich cyclocarya paliurus and zymoyic fungal powder of cordycepes sinensis compound preparations. |
