Timosaponins are the main active ingredient of Rhizoma Anemarrhena commonly used traditional Chinese medicine.According to the relevant Chinese medicine records,Timosaponin B-Ⅱ has been reported to inhibit platelet aggregation,scavenge free radicals,reduce blood lipid,anti-tumor and improve cognitive function and the ability of learning and memory function.In the present study,Timosaponin B-Ⅱ was found that it can protect against hydrogen peroxide cytotoxicity in vitro.Furthermore,the possible mechanism was investigated,which of action involved in the neuroprotective effect of TB-Ⅱ against peroxide-induced injury in PC12 cells.The results showed that Timosaponin B-Ⅱ possessed potent antioxidant activity,so that pre-treatment with TB-Ⅱ prior to H2O2 exposure significantly increased cell viability and reduced the release of LDH.Moreover,Timosaponin BII reduced intracellular accumulation of ROS and inhibited apoptosis.We then employed a metabonomic method based on ultra performance liquid chromatography coupled to mass spectrometry to explore global metabolic characters of PC12 cells in a hydrogen peroxide cytotoxicity model in vitro.We found 35 metabonomics with significantly difference in model group.In summary,our study has investigated and explained the essence of the possible mechanism of the neuroprotective effect of TB-Ⅱ against peroxide-induced injury in PC12 cells by metabolomics method and molecular biology values,which shed a new light to the treatment for oxidative stress-derived neurodegenerative disorders.
