1.Research on PRP influencing the skin structure,barrier cornified envelope development of newborn suckling mouseBackgroundAs the largest human organ,skin is composed of cuticular layer and dermal layer,it is the first barrier against the damages from external environment.Speaking of the barrier function of skin,stratum corneum(SC)is the major barrier that prevents the exogenous stimulations and antigens from entering the organism and the internal water-electrolyte form being lost.The cornified envelope(CE)formed inside the stratum corneum is the foundation of skin barrier.The infants could reach the same skin structure with adults completely at the age of about three years old,in the meantime,the lower skin barrier function is easy to arouse the endogenous immune reaction and skin injury in case of exogenous stimulations to evoke a series of diseases related to the barrier function disorders of skin.According to the clinical report,the pathogenesis of various skin diseases is related to the functional declines and disorders.Research on skin barrier is increasingly noted and therapeutic tools of enhancing the barrier function of skin have become the key pints to prevent and cure various skin diseases.Due to the rich growth factors etc.,PRP can not only improve the water content of skin but also strengthen the immune protection capability of skin against the external world,reduce the stimulations to skin from the external objects and restore the skin to the optimum state.Therefore,the newborn C57BL/6 suckling mouse model is adopted in this research to observe the PRP’s intervention in the development process of skin structure and cornified envelope.ObjectiveTo explore the influences of PRP during the hair follicle development process of newborn nursling mouse.MethodsTake 40 newborn C57BL/6 suckling mice and group them randomly into experimental group with PRP and control group with normal saline,20 mice for each group.In the central area of back,the 0.05ml platelet concentration of the experimental group is10~9/ml PRP with intradermal injection on single point and the control group is injected with equivalent normal saline.Take 5 animal samples respectively from each group at 3d,5d,7d and 10d for the paraffin-embedded routine HE of full-thickness skin tissue,Masson staining to inspect the interest protein of immunohistochemistry technique,make histological observation and shooting on the tissue slice,adopt the ImageJ software to measure the grey level of skin and IOD value expressed by interest protein,the ipp software to measure the thickness of dermal layer and cuticular layer,the spss statistical software to make analysis and calculation on the data.ResultPRP has significantly improved the development process of cuticular layer and dermal layer in the skin tissue structure of newborn suckling mouse.Also,the PRP intervention therapy has promoted the early cutin to form the cellular multiplication and differentiation;in the meantime,the PRP intervention therapy has escalated the expression levels of early Ioricrin and Involucrin of the skin tissue for the newborn suckling mouse to a marked degree.ConclusionPRP plays a significant role in promoting the growth of early cuticular and dermal structures of the suckling mouse as well as improving the barrier function of skin to be mature and complete.2.Research on PRP influencing the hair follicle development of newborn suckling mouseBackgroundAs the largest human organ covering the body surface,skin is the natural coat of human body that directly contacts with the current external environment,hence extremely important to maintain the homeostasis.It has multi-functions such as barrier,absorption,feeling,secretion and excretion,body heat regulation,substance metabolism,immunization and so on.Hair follicle is one of the important accessory organs of skin,the epidermal stem cells on its isthmus are capable of providing cell source for the epidermal components of skin in the physiological status and trauma repairing.Lipsotrichia or psilosis is a common and incurable disease that can appear in the early life;there are limited options for its therapeutic schedule.In addition,people are expecting a rather perfect restoration for the trauma repairing of skin tissue or regenerative repairing of hair surface;in which,research on the development process of skin tissue and its accessory organs of hair follicle is increasingly noted.PRP secretes various growth factors,cell factors and other active products.According to the clinical research,PRP can be used for skin trauma repairing to relieve pains and reduce scars.Also,PRP is more and more important in research on the skin barrier,metabolism of pigment,accessory organs such as sweat gland and hair follicle.In this research,animal models of the newborn C57BL/6 suckling mouse are adopted to intervene the development process of hair follicle with PRP.ObjectiveTo explore the influences of PRP during the development process of hair follicle of the newborn suckling mouse.MethodsTake 40 newborn C57BL/6 suckling mice and group them randomly into experimental group with PRP and control group with normal saline,20 mice for each group.In the central area of back,the 0.05 ml platelet concentration of the experimental group is 109/ml PRP with intradermal injection on single point and the control group is injected with equivalent normal saline.Take 5 animal samples respectively from each group for gross observation at 3d,5d,7d,10 d and cut the paraffin-embedded routine HE of full-thickness skin tissue,staining to make histological observation and shooting on the tissue slice;adopt the Image J software to measure the grey level of skin and the spss statistical software to make analysis and calculation on the data.ResultAccording to the gross observation at 3,5,7d,the changes in color during the skin development of mouse in the PRP group appear earlier than the control group;the statistical result of its skin grey level inspection is roughly consistent with the gross observation(P≤0.01),what’s more,according to the gross observation,at 10 d,the hair density of mouse in the PRP group is significantly higher than the control group.According to the HE staining result,the number and development state of skin tissue of mouse in the PRP group on the same day are obviously superior to the control group.Conclusion PRP can effectively promote the development and maturation process of hair follicle.
