Backgroud: Bladder cancer is the most common manlignancy in the urinary system and one of the top ten manlignancies.The SOX30 gene has been shown to play a tumor suppressing role in lung cancer,but its role has been clearly reported.Objectives: To detect the expression level of SOX30 gene in human bladder cancer tissues and cell lines;To explore the relationship between the expression level of SOX30 gene and the clinicopathological features of bladder cancer;To study the relationship between the expression level of SOX30 gene and the clinical prognosis of bladder cancer;To explore the effect of SOX30 gene expression on the biological function of bladder cancer cells.Methodolog: q RT-PCR was performed to examine the m RNA expression of SOX30 in 30 pairs of bladder cancer and pair-matched adjacent normal bladder tissues,4 bladder cancer cell lines.The protein expression of SOX30 in BC tissue and cell lines was evaluated via western blot and immunohistochemistry.In addition,the clinical and prognostic significance of SOX30 in BC were assessed using a Kaplan-Meier analysis.Furthermore,we measured cell proliferation,cell migration,cell invasion,and cell apoptosis by means of a cell counting kit-8(CCK-8)assay,transwell assay,and flow cytometry,respectively.Resμlts: Expression levels of SOX30 were markedly lower in BC cells and tumor tissues than those in adjacent noncancerous tissues.Moreover,clinicopathological analyses showed that low SOX30 expression was positively related to an advanced tumor,node,and metastasis(TNM) stage(P=0.019),but there was no significant correlation between and age(P=0.093),sex(P=0.639),tumor histological grade(P=0.068),Tumor size(P=0.150)and Pathological type(P=0.317),Furthermore,low SOX30 expression significantly correlated with a reduced survival rate(P=0.0388).Functional analyses revealed that SOX30 overexpression attenuated cell proliferation,invasion,and migration,while promoting apoptosis in BC cells.Conclusion: The present study shows that SOX30 is downregulated in BC tissues and is related to a poor prognosis and advanced TNM stage.Furthermore,our findings demonstrate that SOX30 plays an important role in the proliferation,invasion,migration,and apoptosis of BC cells,suggesting that SOX30 may act as a tumor suppressor gene and might be a novel and promising candidate to improve the prognosis and therapy for BC.
