Studies On The Activity Of β-amyloid Bifunctional Peptide And Screening Of Tau Single Chain Antibody

Alzheimer’s disease(AD),the most common cause of dementia,is a neurodegenerative disease in central nervous system with complex mechanism.It affects the memory,learning ability,and judgment of patients.Advanced Patients can’t care for themselves and need special care,bring a great financial burden to their families and society.The core pathological hallmarks of Alzheimer’s disease in the brain are amyloid plaques consists of β-amyloid outside neurons,neurofibrillary tangles consists of Tau inside neurons,neuronal apoptosis and brain atrophy.Here,we investigated the dual role of a mimotope of Aβ oligomers and the screening of tau specific scFv,briefly described as follows:1.A mimotope of Aβ oligomers may also behave as a beta-sheet inhibitorwe obtained a mimotope of Aβ oligomers,AOEP2,by screening a peptide library using oligomer-specific antibodies.The antibodies induced by AOEP2 specifically recognize Aβ oligomers rather than monomers and fibrils.Interestingly,the AOEP2 peptide binds to Aβ monomers and inhibits the formation of Aβ oligomers and β-sheet structure,reduces Aβ42-induced neurotoxicity and decreases the release of proinflammatory cytokines.Taken together,AOEP2,a novel multifunctional peptide directly or indirectly targeting Aβ,has promising therapeutic potential for AD.2.Screening Tau oligomers specific single chain antibodies using phage display technologyThe single-chain antibodies(scFv)can be screened from commercial phage antibody libraries using Tau oligomers as a target by phage display technology.We changed the target protein concentration and washing conditions during four rounds of panning process,enriching the target scFv at last.Then we detect the binding ability by the phage ELISA.Finally,detect the binding ability of the purified scFv.
