[Background]Procollagen lysine,2-oxoglutarate 5-dioxygenase(PLOD2)is a functional enzyme present in the cytoplasmic rough endoplasmic reticulum,translational modification of proteins,fibrosis,and tissue remodeling.Studies have shown that PLOD2 and tumor progression,prognosis,metastasis and recurrence are closely related.PLOD2 overexpression is associated with poor prognosis of glioma such as liver cancer,bladder cancer and cervical cancer,which may be the prognostic indicator of tumor.However,the correlation between the expression of PLOD2 in laryngeal carcinoma and the prognosis of laryngeal carcinoma is not clear.Therefore,this study collected clinical laryngeal cancer tissues and paracancerous tissues,detected the expression of PLOD2 using immumohistochemistry,RT-PCR and Western blotting,and analyzed the prognostic factors of laryngeal cancer using statistical methods such as sturvival analysis and regression analysis Factors,which in turn provide experimental evidence for diagnosis and prognosis of laryngeal cancer.Expression of PLOD2 in laryngeal carcinoma and its adjacent tissues[Objective]The study measured the expression of PLOD2 in laryngeal carcinoma and laryngeal squamous cell carcinoma tissues and provided experimental evidence for the diagnosis and treatment of laryngeal carcinoma.[Methods]Collected in Guangdong Provincial People’s Hospital ENT 2008 to 2015 clinical diagnosis of laryngeal cancer in patients with surgical specimens of 114 cases,all specimens have corresponding normal tissue next to cancer.All tissue specimens were fixed with formaldehyde and paraffin embedded.Immunohistochemistry,PCR,Western Blot and other methods of PLOD2 expression in laryngeal cancer and laryngeal cancer tissue qualitative and quantitative studies.[Results]1,PLOD2 protein in the laryngeal cancer tissue was positive for immunostaining brown particles,mainly in the cytoplasm;the main color for the different shades of brown,and the clinical stage later,the more the number of particles the darker the color.2,There were significant differences in PCR between laryngeal carcinoma and adjacent normal tissues(p<0.05).The PCR of laryngeal carcinoma was higher than that of adjacent normal tissues.3,The difference of the protein expression in laryngeal carcinoma and adjacent normal tissue was statistically significant(p<0.05),the expression level of protein in laryngeal carcinoma was higher than that in adjacent normal tissues.[Conclusion]The expression of PLOD2 in laryngeal carcinoma tissues was higher than that in adjacent normal tissues.Correlation between PLOD2 and prognosis of laryngeal cancer[Objective]To study the correlation between PLOD2 and the prognosis of laryngeal cancer and to provide a theoretical basis for the prognosis of laryngeal cancer.[Methods]Collected in Guangdong Provincial People’s Hospital ENT 2008 to 2015 clinical diagnosis of laryngeal cancer in patients with surgical specimens of 114 cases,all specimens have corresponding normal tissue next to cancer.All tissue specimens were fixed with formaldehyde and paraffin embedded.Survival time from the date of diagnosis of the disease until the end of follow-up date,or due to tumor recurrence,metastasis and other causes of death of the patient’s date.Collection of clinical specimens of laryngeal cancer pathological parameters(such as:clinical stage,TNM classification,etc.)statistics.Survival analysis and cox regression analysis and other statistical methods to analyze the PLOD2 prognosis of laryngeal cancer.[Results](1)The level of POLD2 protein expression was correlated with clinical stage and T stage,but not with N stage and M stage.(2)Survival analysis of laryngeal cancer patients showed that the 3-year and 5-year survival rates of patients with low expression of PLOD2 protein were 93.01%;3-year and 5-year survival rates of PLOD2 protein high expression patients were 70.65%and 65.11%Survival rate was statistically different.(3)Univariate Cox regression analysis showed that M staging and PLOD2 protein expression(p = 0.002),clinical stage and T stage were the risk factors affecting the growth of laryngeal carcinoma.Multivariate Cox regression analysis of risk factors with statistically significant growth of laryngeal tumors showed that PLOD2 protein expression and M staging are highly correlated with poor prognosis in laryngeal cancer patients.[Conclusion]PLOD2 protein expression directly affects the survival rate of patients,and the expression level of PLOD2 protein is highly correlated with the poor prognosis of patients with laryngeal cancer,and may become a new molecular marker for the prognosis of laryngeal cancer. |
