As we all know,the liver cancer is a very common type of malignant tumor with high recurrence and high metastasis,which has great harm to people’s life and health.Development of HCC is a multi-step and multi-factor process.Therefore,we need to find out that new drugs can effectively inhibit the metastasis of liver cancer and thus can effectively control the patient’s condition.Traditional Chinese medicine plays a very important role in the prevention and treatment of liver cancer as our country’s traditional medicine.Scutellaria javanica Jungh is a plant of Scutellaria labiaceae.However,the effects on chemical and pharmacological about the Scutellaria javanica Jungh remain unclear.From the previous experiments,flavonoids richly exist in Scutellaria javanica Jungh and have been screened for activity.Previously,a new flavonone,Scutellaria javanica Jungh baicalein(5,6,2’-trihydroxy-7-methoxy flavanone)in Scutellaria javanica Jungh,which was found to have anti-tumor effect and induce the apoptosis of HepG-2 cells.However,the mechanism of its antitumor action is not clear.It is necessary to carry out in-depth and systematic research on the role and mechanism of the anti-tumor of the flavonone from Scutellaria javanica Jungh,whether about basic or applied research,especially the study on the mechanism of liver cancer Scutellaria javanica Jungh.The aim of the present study was to investigate the effects of Scutellaria javanica Jungh baicalein from Scutellaria javanica Jungh on the invasion and migration of HepG-2 cells and its possible mechanism.In this research,Transwell chamber was used to detect the changes of the invasion and migration ability when HepG-2 cells were treatment with Scutellaria javanica Jungh baicalein.Western blot was used to detect the protein expression of the molecules(MMP-2,MMP-9 and E-cadherin)associated with invasion and migration.In addition,the protein expression of key molecules about PI3K/AKT and MAPK/ERK signaling pathway,including AKT,p-AKT and p-ERK1/2 were evaluated by Western blot assay as well.In conclusion,the invasion and migration ability of HepG-2 cells were inhibited with the increase of drug concentration in a dose-dependent manner.It indicated that Scutellaria javanica Jungh baicalein could inhibit the invasion and migration of HepG-2 cells.Expression invasion and migration regulatory factors are altered:MMP-2 and MMP-9 are down-regulated,but expression of E-cadherin protein is enhanced by drug treatment.Furthermore,Western blot assay was used to measure the expression of p-AKT and AKT in PI3K/AKT signaling pathway as well as the expression of p-ERK1/2 in MAPK/ERK signaling pathway.The results showed that Scutellaria javanica Jungh baicalein could suppress PI3K/AKT and MAPK/ERK signaling pathway,as well as p-AKT and p-ERK1/2,but AKT was basically unchanged.Our datas indicated that Scutellaria javanica Jungh baicalein could effectively inhibit the activation of PI3K/AKT and MAPK/ERK signaling pathway.To sum up,Scutellaria javanica Jungh baicalein can inhibit the invasion and migration of HepG-2 cells in a dosage-dependent manner.Scutellaria javanica Jungh baicalein can affect the invasion and migration of tumor related genes,thereby inhibiting the invasion and migration of tumor cells,and its mechanism may be through inhibition of PI3K/AKT and MAPK/ERK pathways,thereby inhibiting the expression of downstream molecules MMP-2 and MMP-9,and promoting the expression of E-cadherin,and ultimately inhibit the invasion and migration ability of HepG-2 cells.
