Objective: To investigate the treatment for xerophthalmia rabbit by using the lacrimal suppository with extended release dosage forms.Methods: A total of 36 male rabbits(36 right eyes),were randomly divided into 6 groups(6 rabbits per group).Group A: hydrogel lacrimal suppository treatment;Group B: nano-silver lacrimal suppository treatment;Group C: nano-silver-graphene lacrimal suppository treatment;Group D: fluorometholone lacrimal suppository treatment;Group E: no treatment group;Group F: control group.According to the existing research methods,the rabbits of Group A to D were made into xerophthalmia rabbit models by benzalkonium chloride eyedrops(BAC,0.1%BAC,right eyes,Bid,7days).The insertion of various 3D-printed lacrimal suppository were made on days 8,respectively.Examination of the Schirmer’s test(SIT),corneal fluorescein(FL)and corneal photographed were performed on day0,day7,day10,day14.Conjunctival impression cytology(CIC),transmission electron microscopy(TEM)and scanning electron microscopy(SEM)technology were apply to analysis of the treatment effect for dry eye rabbit by lacrimal suppository with extended release dosage forms by contrast.Result: Compared with the control group,The scores of FL were significantly increased afteron days 7(P=0.003,P<0.05).CIC examination showed that the number of conjunctival goblet cells decreased and the nuclear were pyknosis and disintegration;corneal HE staining showed most corneal epithelial were damage,shedding,disorder,the layers of epithelial cell were increased and goblet cells decreased;TEM examination indicate that the surface of corneal epithelial cell were irregular and shedding,the microvilli were fracture,disorder,nuclear gap were expanded,the number of desmosomes and hemidesmosomes contacts significantly reduced,the number of mitochondria in corneal stroma layer were increased and edema.After lacrimal occlusion with various 3D-printed lacrimal suppository,the examinations of SIT were significantly relieved in group D(P=0.006,P<0.05),the scores of FL were significantly decreased(P=0.005,P<0.05).CIC examination showed that the number of conjunctival goblet cells were significantly decreased(P=0.008,P<0.05).Corneal HE staining showed the corneal epithelium become completer,intercellular desmosomes contacts are closely connected,the number of goblet cells increased;results of TEM examination showed that the microvilli of corneal epithelial become thicker,neat,reduced mitochondria edema.Conclusion: Lacrimal suppository with extended release dosage forms,are effective for the xerophthalmia.
