| The human α/β-hydrolase domain 16A(ABHD16A)also called human leukocyte antigen-B associated transcript 5(BAT5)is one of the ABHD protein family members and highly conserved in mammals.Because the ABHD16 A gene is located in the major histocompatibility complex(MHC)III gene cluster,it is speculated that it may participate in the immune regulation of the body,however,the study of its function has been less reported so far.Previous studies of MHCIII interacting proteins revealed that ABHD16 A interacts with interferon-inducible transmembrane protein 1(IFITM1).IFITM1 belongs to the IFITM family of proteins.This family of proteins not only has antiviral activity,but also plays an important role in cell proliferation and migration,cell adhesion,and signal transduction.A previous study of laboratory showed that IFITM-/-mice developed an inflammatory reaction,and in the liver of mice,the protein content of ABHD16 A increased significantly.It is speculated that the interaction between ABHD16 A and IFITMs has a certain relationship in the inflammatory response,antiviral effect and other functions,and conduct research accordingly.The human ABHD16 A and IFITM1,IFITM2,and IFITM3 were used as research objects.The yeast two-hybrid system and subcellular colocalization were used to study the interaction between ABHD16 A and IFITM.The gene was transiently over-expressed and LPS stimulated to explore its role in inflammation.The effect was combined with the detection of ABHD16 A and IFITMs expression in blood samples from clinical hepatitis B patients.The results showed that yeast two-hybrid assay demonstrated the interaction between ABHD16 A and IFITM1.Confocal laser scanning microscopy showed that ABHD16 A was colocalized with IFITM1,IFITM2,and IFITM3 in the cell membrane region.IFITM1,IFITM2 and IFITM3,the expression of IL-6 was decreased,the expression of anti-inflammatory IL-10 was increased(P<0.05),and the levels of pro-inflammatory cytokines INOS and TNFα were also increased after overexpression of ABHD16 A.After stimulated by LPS,the m RNA levels of ABHD16 A,IFITM1,IFITM2,IFITM3,and TNFα were up-regulated(P<0.05),indicating that ABHD16 A,IFITM1,IFITM2,and IFITM3 play an important role in the inflammatory response.The results of blood samples from patients with clinical hepatitis B showed that the expression of ABHD16 A was down-regulated,the expression levels of IFITM1,IFITM2,and IFITM3 in Dasanyang patients were down-regulated,and the expression levels of IFITM1,IFITM2,and IFITM3 were up-regulated in patients with Xiaosanyang,suggesting that the changes in the expression of ABHD16 A,IFITM1,IFITM2 and IFITM3 caused by hepatitis B virus are worth further investigation.This study demonstrated the interaction between ABHD16 A and IFITMs,and conducted a preli minary study of their expression in inflammatory reactions and hepatitis B patients,providing the theoretical basis for further research on the functional aspects of ABHD16 A and IFITMs in inflammation and anti-hepatitis virus. |
