| The plant is a genus of herbs of Crinum Crinum red spider lily plant,red spider lily petals white,a faint fragrance.Crinum plant contains a variety of essential oils,alkaloids,flavonoids,polysaccharides,fatty acids and other chemical components.Its heel leaves can be used as medicine.The anti-inflammatory,anti choline-like,and treatment of cardiovascular and cerebrovascular diseases have a rich pharmacological effect.At present,research on the volatile components in the leaves of Crinum less of its anti-tumor effect has not yet in-depth study.This project uses dry Crinum leaves,extract the volatile components,and determine the content of main active substances by extraction and GC-MS analysis method for the study of its volatile components.In order to further research on the anti-tumor Crinum provide effective scientific basis.This project uses steam distillation,extraction,distillation and extraction of three kinds of methods for extracting volatile constituents in leaves of crinum dry.Through orthogonal test,we can see that the best extraction process is to select the best extraction process,namely,the extraction time is 120min,the extraction temperature is 70 degrees,and the extraction time is 6 hours.The extraction rates of the three extraction methods were 0.55%,0.95%and 0.72%respectively.GC-MS was used to screen the best heating conditions at 75 C and 1.5 min at constant temperature.The temperature was heated to 100/min at 5/min,and maintained at 0.5 min.The temperature was increased from 15/min to 130 C,and 4 min was maintained.The temperature increased to 5 degrees/min to 160 degrees centigrade,maintaining 10 min,and increased to/min temperature at/min temperature.Keep 15 min.Respectively 15 and 14 compounds detected by steam distillation and distillation extraction method,containing 18 components in the volatile oil extraction method,according to the detected content found three kinds of volatile oil in the proposed method can detect the main component of linoleic acid,the linoleic acid anti-tumor activity further research on screening.Using the method of CCK-8,detection of Crinum proliferation inhibition effect of total volatile oil constituents on tumor cells,using five concentration experiment,using SPSS orthogonal test to calculate the LD50,that in Crinum total volatile oil inhibited the proliferation of HepG-2 cells,whereas the total volatile oil components of linoleic acid was highest,that of linoleic acid inhibit the proliferation of HepG-2 cells,then remove the linoleic acid repeating the experiment,determination of linoleic acid also inhibited the proliferation of HepG-2,showed a linear relationship with the concentration increased.Using an inverted microscope and fluorescence microscope was used to observe the morphology of cells to give medicine,found the change after administration of cell shape,increasing apoptosis,cell edge wrinkling,foaming phenomenon,the greater the concentration of apoptosis phenomenon is more obvious,linoleic acid on HepG-2 cell proliferation inhibition with the same.Finally,using the test for the rate of cell apoptosis of drug flow cytometry,positive group apoptosis rate was(48.503+0.09)%,blank group apoptosis rate was(1.099+0.15)%,the high concentration group apoptosis rate was(36.155+0.02)%in the concentration group apoptosis rate was(22.525 +0.12)%.The low concentration group the apoptosis rate was(18.433+0.11)%.Study on the volatile components in the leaves of Crinum found that the volatile components in the leaves of Crinum could inhibit the proliferation of tumor cells.The composition of linoleic acid may be the main component of its anti-tumor effect. |
