| Objective: Based on the relationship between heart and brain,the differential protein and the Naotaifang interaction mechanism of cerebral infarction were explored by proteomics,and the molecular mechanism of cerebral infarction prevention and treatment by Naotaifang was preliminarily clarified from the contact of heart to brain.Methods: 1.Based on the physiological and pathological links between the heart and brain,the common molecular targets and signal transduction pathways of the ischemic diseases in the heart and brain are searched by using the bioinformatics database.2.Using the method of network pharmacology to explore the target and signal transduction pathway of the effective components of the Nataifang,and to compare the molecular targets and signal pathways of cerebral infarction and myocardial infarction,and to find their common targets and signal pathways.3.The MCAO model of SD rats was established,which were divided into sham operation group(S),sham operation + Naotaifang Group(SN),model group(CI),model + Naotaifang Group(CN)and edaravone group(CE).TMT protein technique was used to find the difference protein of plasma in each group,and the bioinformatics were used to analyze CN/CI,SN/S,CN/S,CN/SN,CI/S.The results of GO analysis and KEEG pathway were found in five contrast groups,and the correlation between heart-brain through plasma differential protein and its pathway was found after cerebral infarction.Results: 1.There are close links between the heart and brain,and 2 common targets for ischemic heart and brain disease: 5-serotonin 2B receptor and plasminogen,5 common signal transduction pathways: calcium signaling pathway,complement and coagulation cascade reaction,gap junction,neuroactive ligand receptor interaction,muscle interaction Kinesin cytoskeleton regulation;2.There is one identical molecular target in the target site of Naotaifang and cardiovascular and ischemic diseases,namely plasminogen;which involves 4 identical signaling pathways,are complement coagulation cascade and neuroactive ligand-receptor interaction,regulation of actin cytoskeleton and calcium signaling pathway.3.In the signal pathway related to the heart,1 involved in SN/S(arrhythmogenic right ventricular cardiomyopathy related pathways)and 3 involved in CN/CI(dilated cardiomyopathy,hypertrophic cardiomyopathy,arrhythmogenic right ventricular cardiomyopathy),and 2 involved in CN/S(dilated cardiomyopathy,hypertrophic cardiomyopathy related pathways),and 3 involved in CI/S(dilated cardiomyopathy,hypertrophic cardiomyopathy,renin angiotensin system related pathways),while CN/SN was not involved.Conclusion: The ischemic disease of the heart and brain involves common molecular target and signal transduction pathway,which confirms the correlation between the heart and the brain,and the cerebral infarction and myocardial ischemia may have a common therapeutic approach,and the cerebral infarction have clear target and signal transduction pathway for the prevention and treatment by Naotaifang,and it is a multi target and pathway.By analyzing the differential proteins in the prognosis of cerebral infarction and drug treatment,it was found that some of the pathways involved were associated with heart disease,indicating that the cardiac related changes were caused after the occurrence of cerebral infarction,and the relationship between the heart and the brain was verified,which may indicate that the target of cerebral infarction is not only limited to the brain. |
