Objective: To explore the protective effect of olaparib on the preservation of liver perfusion in rats at different temperatures.Methods: 80 male rats in Wistar were selected.The 50 rats were randomly divided into 5 groups: G0:the control group(non-instillation and preservation of the liver)10;G1:the hypothermia group(only UW solution perfused at 4 ℃)10;G2: Low temperature joint ora panitan group(ora panitan joining UW solution 4 ℃ perfusion of liver)10;G3:The normal temperature group(37℃ pure UW solution perfusion liver)10.G4;normal temperature joint Olaparib group(Olaparib joining UW solution 37 ℃ perfusion of liver)10;The other 30 were liver transplant recipients.rats preoperative fasting 12 h,water,preoperative with 10% chloral hydrate in 0.3 ml / 100 g via abdomen anesthesia,after anesthesia with abdomen "ten" glyph incision through into the abdominal cavity,the separation of the portal vein,indwelling needle inserted into the portal vein after take out the core fixed with 4 a thread of silk,connected with mechanical pump,with(6.0±1.0)ml/min flow pumping fluid of PBS,and inferior vena cava under cut liver bleeding continuous lavage in situ liver at room temperature,20 min after liver into uniform khaki,perfusion effluent is clear after the complete separation of the liver.The G0 group of isolated livers were directly examined for liver biopsy.The G1 group of isolated liver was placed in a beaker containing 200 ml UW solution,and the beaker was placed in crushed ice to maintain the perfusion solution at about 0-4℃.The mechanical pump inlet was placed in a beaker,and the G2 group was placed in vitro.Into a 200 ml UW solution containing olaparib(concentration 50 nM)was placed in crushed ice to maintain a perfusion solution of approximately 0-4℃.The G3 group of isolated livers was placed in a beaker containing 200 ml of UW solution,and the beaker was placed in a constant-temperature water bath to maintain the perfusate at 37℃.The G4 group of isolated liver was placed in a 200 ml UW solution containing olaparib(concentration: 50 nM),and the beaker was placed in a constant-temperature water bath to maintain the perfusion liquid at about 37℃.The above G1-G4 groups kept refluxing for 4h and then withdrew the mechanical pump and continued to save at 0-4℃ for 4h.Four of all groups were fixed to take the left hepatic lobe about hepatic soybean size HE and TUNEL staining,and the expression of caspase-3 was observed by immunohistochemistry.All cytokines such as Bax,Bacl2,caspase-3,caspase-9,IL-1β,IL-6,TNF-α in liver tissue were extracted and compared.The orthotopic liver transplantation was performed on the livers of 5 rats in each group to observe the postoperative 2-day survival rate to evaluate the effect of perfusion preservation in each group.Six orthodontic liver transplants were performed in 6 rats in each group.Liver function was observed on the 1st,3rd,and 7th postoperative day to evaluate the effect of perfusion preservation in each group.Results:1.The perfusion and preservation of perfusion at room temperature was more effective than that of cryopreservation alone.The apoptosis and inflammation were decreased in donor liver tissue.2.The addition of olaparib in UW solution could reduce the apoptosis and inflammation in donor liver tissue.3.Combined Olaparib perfusion preservation of donor liver can better reduce apoptosis and inflammatory response.4.Normal liver function combined with olaparib was better for liver preservation after liver transplantation.Conclusions:1.Perfusion preservation at room temperature is superior to cryopreservation;2.Olaparib is beneficial for hepatic perfusion preservation;3.The perfusion effect of normal urinazuron combined with Olaparib UW solution is more obvious.4.It is better to infuse liver transplantation after preservation of donor liver at room temperature with olaparib.
