Polycystic ovary syndrome(PCOS)is an endocrine disorder syndrome,in which reproductive dysfunction and abnormal glucose metabolism coexist.50%-70%of PCOS patients have IR.The metformin,a commonly used insulin sensitizing drug,has a therapeutic effect on metabolic and reproductive dysfunction,and can increase the oocyte quality of PCOS mouse models induced by dehydroepiandrosterone(DHEA).More studies have showed that chronic inflammation was found in patients with PCOS,while chronic inflammation can mediate insulin resistance,and insulin resistance of PCOS can increase androgen production and induce premature maturation of granulocyte maturation,impair the biological effects of granulosa cells,and influence the growth and development of follicles,thereby affecting the growth and development of oocytes.Correspond to this,Pachymic acid,a triterpenoid compound is naturally occurring in Chinese herbs such as Poria cocos and Ganoderma lucidum,with anti-inflammatory antioxidant,hypoglycemic and other pharmacological effects.Therefore,this experiment aims to compare metformin,to study whether pachymic acid can prevent inflammation and alleviate insulin resistance,inhibit the production of androgen,improve the ovary microenvironment,and effectively inhibit the damage of granulosa cells,thus improving impaired oocyte quality of mouse with polycystic ovary syndrome.In this study,we used PCOS mouse model induced by the DHEA to investigate the effects of pachymic acid on metabolic abnormalities and reproductive dysfunction.Changes in body weight,glucose tolerance,and glucose metabolism indicate that pachymic acid can reduce insulin resistance in PCOS mice.Tests of IL-6 and TNF-a protein and serum factor levels indicate that pachymic acid can partially inhibit the production of inflammation.The detection of sex cycle and hormone levels in PCOS mouse showed that pachymic acid had a weak effect on hormones.Detection the morphology and apoptosis of ovarian,oocyte and embryo revealed that pachymic acid can improve ovarian function,improve oocyte quality and late development.Our data indicate that pachymic acid can inhibit inflammation and alleviate insulin resistance in PCOS mice,thereby inhibiting granulosa cell damage,inhibiting oocyte oxidation and apoptosis,and indirectly increasing the oocyte quality of mice with polycystic ovarian syndrome.
