Objective:1.The resources of Akebia quinata in Anhui province were investigated,and the resources distribution of Akebia quinata in Anhui province were mastered,and the climatic environment was preliminarily analyzed.2.The fingerprints of Akebia quinata were established in Anhui province to provide basis for establishing the quality control standard of Akebia quinata.3.The analytical methods are established for Akebia quinata in different habitats and seasons,and to understand the quality and suitable harvest season of Akebia quinata in Anhui province.4.The distribution of three terpenoid saponins in different tissues and the characteristics of accumulation and change in different diameters were clarified through the histochemical study and the content detection of effective components in different diameter and different parts of Akebia quinata.Methods:1.Geographic Information System(GIS)was used to analyze the resources of Akebia trifoliate,and the climatic environment of Akebia quinata was analyzed through a variety of climatic indicators.2.The fingerprint of Akebia quinata leaves from different habitats was established by ultra performance liquid chromatography.3.The content of effective components in Akebia quinata leaves from different habitats and seasons was detected by ultra performance liquid chromatography.4.Using the method of vanillin glacial acetic acid and perchloric acid reagent coloration,the effective components of three terpenoid saponins in Akebia quinata were located,and the content of three effective components in different tissue structures and different diameters of five leaves were determined by high performance liquid chromatography.5.The chemical constituents of Akebia quinata were qualitatively analyzed by ultra performance liquid chromatography / quadrupole tandem time-of-flight mass spectrometry.Results:1.Through the result of the resource survey,the distribution map of the resources of Chinese medicine wood Tong in Anhui province was constructed by geographic information system.It was found that three species of wood are mainly distributed in the hilly area of Jianghuai,Anhui and Dabie Mountains,and the wild resources are rich in the southern Anhui and Dabie mountains.It is the most abundant in the hilly areas of the Yangtze and Huaihe rivers.The principal component analysis of climate indicators in Anhui Province found that the annual average temperature,the average temperature of the most cold month,the extreme minimum temperature,the biological temperature,the Xu Wenduo humidity index,the possible evapotranspiration rate and ?10 ? accumulated temperature,the 7 climate factors have the greatest impact on the local climate and environment.2.The fingerprint of 18 batches of Akebia quinata in Anhui province was established.The precision,stability and repeatability of the experiment were investigated.The results showed that the fingerprint of Akebia quinata was feasible,and the fingerprint atlas was in accordance with the technical requirements of the establishment of the fingerprint of Chinese medicine.20 common peaks were determined according to the relative retention time,and the similarity calculation of the chromatograms was carried out.The results of similarity calculation of 14 batches of samples were above0.90,and the similarity calculation results of the 4 batch of samples were above 0.80.The results showed that the quality of Akebia quinata in all parts was near,and the relative peak surface of the samples was calculated by cluster calculation.The accumulated Euclidean distance can be divided into two groups of 18 batches of samples: the central and western regions of the Huaihe Huaihe hills and the Dabie Mountains,the eastern part of the Jianghuai Hilly Region and the southern Anhui mountains.The principal component analysis found that the quality of phenethonoside B and oleanolic acid have an important influence on the quality of Akebia quinata.3.The quality of Akebia quinata in different habitats and seasons was analyzed with alpha-Ivy saponins,phenethyl alcohol glucoside B and oleanolic acid.It was foundthat the content of phenethyl alcohol glucoside in Akebia quinata of different habitats increased in direct proportion to the harvest time,but there was no obvious change in alpha-Ivy saponins and oleanolic acid.4.The results of histochemical localization showed that the cortex cells of the cane were dyed purple red,the cork layer and phloem cells were dyed red,the middle column and xylem cells were dyed orange,and the color reaction of the crystal cell layer and the pulp did not occur.Color changes from deep to shallow are: cortex,cork layer,secondary xylem.The content of phenethyl alcohol glucoside B increased with the increase of diameter,and the content reached the highest at 0.9 cm,and then decreased with the increase of diameter.The content of ivy and oleanolic acid is inversely proportional to the diameter of Akebia quinata.The content of 3 effective components in secondary xylem,secondary phloem and phloem layer showed high to low order.5.By establishing a chemical composition database of Akebia quinata and combining with UNIFI analysis software,each molecular ion peak in the total ion flow chart of the anions was screened and identified,and the possible structures of the 38 peaks were determined,of which 32 were three terpene or three terpenoid saponins.Summary:The leaves of Akebia quinata are distributed in all parts of Anhui Province,especially in Jianghuai Hilly Area.At the same time,it is found that the Anhui province can be divided into two types,namely,the Midwest,the Dabie Mountains,the east of the Huaihe Huaihe hill and the south of Anhui Province,especially in the hilly areas of the Yangtze and Huaihe Huaihe hills,are more stable and have good quality.The study found that the best quality of Akebia quinata is 0.9 centimeters in diameter.
