Part 1: Expression of HIPK3 protein in non-small cell lung cancer and its relationship with clinicopathological parameters.Objective: To investigate the expression and biological significance of HIPK3 in non-small cell lung cancer(non-small cell lung cancer,NSCLC)tissues and para cancerous tissues.Methods: Immunohistochemistry was used to detect the expression of HIPK3 in NSCLC tissues and adjacent tissues,and to analyze the relationship with clinicopathological parameters and five-year survival rate.Results: To compared with the adjacent tissues,the expression of HIPK3 was significantly reduced in NSCLC cancer tissues(P <0.001),and the decreased expression of HIPK3 was closely related to the five-year survival rate of the patients(P<0.01).Conclusion: HIPK3 may be a marker for the progression and prognosis of NSCLC patients.Part 2: The regulatory mechanism and biological significance of Mi R-224 to HIPK3 of non-small cell lung cancer cell line A549.Objective: To investigate the regulation of miR-224 on HIPK3 in NSCLC cell line A549 and to analyze its effect on the proliferation and invasiveness of tumor cells.Methods: Using Biological Analysis Prediction,real-time fluorescence quantitative polymerase chain reaction,Western Blot and other mothods to detect the regulation of miR-224 on HIPK3.The effect of miR-224 Overexpression or interference on tumor cells proliferation and invasion ability was detected by MTT,Transwell and cell scratch assay.Results: Mi R-224 could directly target HIPK3 and inhibit the expression of HIPK3,thereby promoting the development of tumor cells.After interfering with miR-224,the proliferation and invasion ability of tumor cells decreased.Conclusion: Mi R-224 affects the proliferation and invasion of NSCLC tumor cells by targeting HIPK3,and provides a new therapeutic target for the treatment of NSCLC. |
