Objective: Invasive breast ductal carcinoma(IDC)is one of the most common histological types of breast cancer.The aim of our study was to investigate the molecular mechanism of IDC and identify its potential therapy targets.In this study,Analysis of invasive ductal breast carcinoma and adjacent tissue microarray data show that the expression of LIPE and COL4A1 relative to cell proliferation and tumor growth of invasive ductal breast cancer.Methods: Gene expression data with accession number of GSE21422 were downloaded from the Gene Expression Omnibus(GEO).They were identified based on limma package of R,and further,Gene Ontology(GO)and pathway enrichment analysis of DEGs were performed through the Database for Annotation,Visualization and Integrated Discovery(DAVID).Protein-protein interaction(PPI)network of DEGs were constructed by STRING database.Survival estimates were computed using the Kaplan-Meier method and differences between survival times were assessed by means of the Log rank test.Results: Totally 108 DEGs were differentially expressed in IDC tissues compared with adjacent non-neoplastic tissues.By comparing the gene lists from GO and KEGG enrichment analysis and PPI network indicated that COMP、PPARG、LEPand LIPE were identified.A significant correlation between lower LIPE expression and higher LIPE expression was identified,and a low LIPE expression was associated to a shorter time to progression.Meanwhile,LIPE mRNA level was lower in IDC tissues than that in normal breast tissues;The total amount of 478 differentially expressed genes in Invasive ductal carcinoma which compared with normal breast epithelial cells were recognized.Functional enrichment analysis proved the most part of differentially expressed genes had connection with ECM-receptor interaction.The two genes lists were contrasted in PPI network,and mi RNA regulation networks,The most two crucial genes were identified in our study,which may be helpful to improve Invasive ductal carcinoma treatment.Additionally,experimental results shows that the COL4A1 gene,one of identified genes,played important roles in both of proliferation and colony formation in Invasive ductal carcinoma.Conclusions: The expression of LIPE was down-regulated between in IDC tissues and adjacent non-neoplastic tissues.Meanwhile,LIPE expression in IDC related to the prognosis of patients and have potential to be biomarkers as diagnostic for breast cancer;Invasive ductal carcinoma could have connection with ECM-receptor mutations.These 9 vital genes could be an important part in the progression of Invasive ductal carcinoma and be offered as therapy targets and prognosis indicator.and the experimental results showed that one of the most crucial genes,COL4A1,was the key gene that influence the proliferation and colony formation of the invasive ductal carcinoma cell.
