Background and objective:Based on the common syndromes of clinical diabetic nephropathy,this study established a diabetic nephropathy rat model of Qi Yin deficiency and blood stasis,prevention and treatment of the rat model by Danzhi Jiangtang capsule,an in-hospital preparation of the first affiliated hospital of our school,which has the characteristics of invigorating qi,nourishing yin and activating blood circulation,changes of Endothelial progenitor cells in Renal tissue of Diabetic Nephropathy Rats,preventive and therapeutic effects of Danzhi Jiangtang capsule on vascular endothelial progenitor cells in renal tissue of diabetic nephropathy rats,Study on the Pathogenesis of Diabetic Nephropathy from the Angle of Vascular Endothelial progenitor cells and the Prevention and treatment Mechanism of Danzhi Jiangtang capsule,It provides a new clue for the prevention and treatment of qi and yin deficiency and blood stasis syndrome in early diabetic nephropathy.Research method:Wistar male rats were fed adaptively for one week,and were randomly divided into normal group and model group.The rats in each group were treated with left nephrectomy and fed with high fat and high sugar for four weeks.Rats were given intraperitoneal injection of low dose streptozotocin in a single dose,and blood glucose was measured by blood test at the tip of the tail.Blood glucose was measured by blood test at tail tip.Blood glucose ≥ 16.7mmol/L was used as model.The rats were randomly divided into three groups: model group,benazepril group,high dose group,middle dose group,low dose group,normal saline group and benazepril group.The traditional Chinese medicine group was given Danzhi Jiangtang capsule in high,medium and low doses by gavage.After 8 weeks of treatment,blood and urine samples were collected,and 24 hours urinary albumin,kidney weight / body weight,blood / urine creatinine,serum superoxide dismutase and malondialdehyde were measured in each group.To observe the pathological changes of renal tissue,take the kidney tissue,make the pathological section,and make the hematoxylin eosin(HE)staining,and observe the pathological changes of the renal tissue.Wester-blot was used to detect the expression of CD34,CD144 and VEGFR-2,CD11 b in renal tissues.Results:1.Glucose:After treatment,blood glucose in rats was significantly higher in group M than in group Z(P<0.05).There was no significant difference between group B and group M(P>0.05).DD group and DZ group were all lower than group M(average P<0.05),the difference was statistically significant;compared with group B,DD group and DZ group DD group were all decreased(P<0.05);DD group was higher than DZ group(P<0.05),DG group was lower than DZ group(P<0.05).2.24 h Urinary albumin24 h urine protein: M group was higher than Z group after modeling(P<0.05),the difference was statistically significant;B group,DD group,DZ group,DG group were lower than M group(P<0.05),difference Statistically significant;Compared with group B,DD group and DZ group were all increased(P<0.05),and there was no difference between group B and DG group(P>0.05);compared with group DG,group DZ,DD Both groups were elevated(P<0.05),and the difference was statistically significant.There was no significant difference between DZ group and DD group(P>0.05).3.Rat serum creatinine and creatinine clearance Rat serum creatinine: Compared with group Z,model building increased(P<0.05),and the difference was statistically significant.Compared with group M,group B,DD group,DZ group,and DG group all decreased(P< 0.05),the difference was statistically significant;DD group was higher than B group(P <0.05),DG group was lower than B group(P <0.05),the difference was statistically significant,there was no significant difference between B group and DG(P <0.05);DD group was higher than DZ group(P <0.05),DG group was lower than DZ group (P<0.05).Creatinine clearance: Compared with Z group,M group was significantly higher(P<0.05);Compared with M group,DD group,DZ group,DG group were all decreased(P<0.05),the difference was statistically significant,M group and There was no significant difference between group B(P>0.05).Compared with group B,the DD group,DZ group,and DG group were all decreased(P<0.05).The difference was statistically significant.The DD group was higher than the DZ group(P <0.05),DG group was lower than DZ group(P<0.05).4.blood malondialdehyde(MDA):Blood malondialdehyde in rats: Compared with group Z,the blood MDA content in group M increased(P<0.05),and the difference was statistically significant;compared with group M,B group,DD group,DZ group,DG group decreased(P<0.05),the difference was statistically significant;DD group was higher than group B(P<0.05),DG group was lower than group B(P<0.05),and there was no significant difference between DZ group and group B(P>0.05);DD group was higher than DZ group(P<0.05),DG group was lower than DZ group(P<0.05).5.Blood superoxide dismutase(SOD):Superoxide dismutase in rats: Compared with group Z,the SOD content in group M was decreased(P<0.05),the difference was statistically significant;compared with group M,group B,group DD,group DZ,group DG were all Elevated(P<0.05),the difference was statistically significant;DD group and DZ group were lower than group B(P<0.05),DG group and B group had no significant difference(P>0.05);DD group Below the DZ group(P<0.05),the DG group was higher than the DZ group(P<0.05).6.Observation of renal histopathological changes by HE staining The renal tissue structure of normal rats is basically normal.There was an increase in glomerular hypertrophy in model set,and there was an increase in mesangial matrix.The mesentery appeared to be thickened and thickened,and nodular hyperplasia was seen.There was a slight change in the high-dose group of Danzhi Jiangtang capsule,only seen in focal glomeruli hyperplasia and partial glomeruli hyperplasia.The nodular hyperplasia in benazepril group and Danzhijiangtang capsule middle dose group was significantly higher than that in Danzhi Jiangtang capsule low dose group,but there was no significant difference between the low dose group and the model blank group.However,no glomerulosclerosis was found in the renal tissues of the rats in each group.7.Elevation of CD11 b in rats :CD11b in rats :Compared with group Z,the expression of CD11 b protein in group M was increased(P<0.05),and the difference was statistically significant.Compared with group M,group B,group DD,group DZ,and group DG were decreased(P< 0.05),the difference was statistically significant;Compared with group B,DG in DD group and DZ group increased(P<0.05),the difference was statistically significant;DD group was higher than DZ group(P<0.05),DG group was lower DZ group(P<0.05).8.Elevation of CD34 in rats :CD34 in rats : Compared with Z group,the expression of CD34 protein in group M was increased(P<0.05),the difference was statistically significant;compared with group M,B group,DD group,DZ group,DG group decreased(P<0.05),the difference was statistically significant;Compared with group B,DG in DD group and DZ group increased(P<0.05),the difference was statistically significant;DD group was higher than DZ group(P<0.05),DG group was lower DZ group(P<0.05).9.Elevation of CD144 in rats :CD144 in rats:Compared with Z group,the CD144 expression in the model group increased(P<0.05),and the difference was statistically significant.Compared with group M,B group,DD group,DZ group and DG group decreased(P<0.05).The difference was statistically significant.Compared with group B,DG in DD group and DZ group was increased(P<0.05).The difference was statistically significant.There was no difference between DD group and DZ group(P>0.05).DG group was lower than DZ.Group(P < 0.05).10.Elevation of VEGFR-2 in rats :VEGFR-2 in rats: Compared with group Z,the expression of VEGFR-2 protein in group M increased(P<0.05),and the difference was statistically significant;compared with group M,group B,group DD,group DZ,group DG The difference was statistically significant(P<0.05).Compared with group B,DG in DD group and DZ group increased(P<0.05),and the difference was statistically significant.DD group was higher than DZ group(P<0.05).The DG group was lower than the DZ group(P<0.05).Conclusion:The experimental study found that the different dosage groups of Dan Zhi Jiangtang granule have a lower effect on blood glucose,urinary albumin and creatinine in rats 、 It can increase the activity of blood superoxide gasification enzyme and decrease the content of malondialdehyde in blood.And to reduce the content of CD34,CD11 b,CD144,VEGFR-2 and so on in vascular endothelial progenitor cells of renal tissue.It can protect kidney function,reduce inflammatory reaction in renal tissue,improve glomerular Mesangial hyperplasia and glomerular fibrosis in diabetic nephropathy rats.Thus delaying the damage of endothelial progenitor cells in kidney tissue of diabetic nephropathy rats.
