Colon cancer ranked as the third most malignant tumor in the world.About 600 thousand people die of colon cancer every year all over the world.Despite the huge threat to human health,the molecular mechanism of colon cancer has not been clearly explained and the mortality rate of it has not been significantly declined.Long non-coding RNA ZEB1-AS1 is first found in hepatocellular carcinoma,and it plays a vital role in proliferation and invasion of hepatoma carcinoma cells.Follow-up researches show it also has an important carcinogenesis effect in other cancers,but there is no report elaborates its function in colon cancer.Our group collected 63 pairs of CRC tissues and paired adjacent non-tumor colon tissues.By qRT-PCR,we found the expression of ZEB1-ASlin colon cancer is significant higher than it in non-tumor tissues.Its expression level is closely related with the clinical pathological features of colon patients.Small interfering RNA(si-RNA)is used to reduce the exoression of ZEB1-AS1,which blocks the cell cycle in G1 phase,reduces the proliferation of colon cells and promotes the apoptosis of colon cells.To explore the molecular mechanism of ZEB1-AS1 in colon cancer,we detected the expression levels of related apoptosis and cycle protein by qRT-PCR.The results showed P15 increased significantly.There was a negative correlation between ZEB1-AS1 and P15,suggesting ZEB1-AS1 is likely to negatively regulate the expression of p15.In order to further explore the relationship between ZEB1-AS1 and P15,our group transfected si-p15 on the basis of si-ZEB 1-AS 1 to down-regulating P15 expression.The result suggested the ability of cell proliferation was rescued,which indicated that P15 is possible to be a target gene of ZEB1-AS 1. |
