The dry rhizoma of Diphylleia sinensis H.L.Li is often used by Tujia medicine to treat diseases like cancer,traumatic injury,rheum arthritis.Flavonoids and polysaccharides from the dry rhizoma of Diphylleia sinensis were mainly studied in this thesis.To reveal the processing mechanism of Diphylleia sinensis by researching the variation of main chemical compositions in crude and processed Diphylleia sinensis.The results showed that the dissolution rate of main chemical components of Diphylleia sinensis increased in different degrees after processing,a few podophyllotoxin could convert into picropodophyllotoxin.The total flavonoids of Diphylleia sinensis(DSTF)were extracted and determined its content by UV method,the results showed that the extraction rate and content of flavonoids both increased in processed products especially in vinegar-stir-baked product compared with crude.The anti-inflammatory and analgesic effects of total flavonoids in the dry rhizoma of processed and crude Diphylleia sinensis were evaluated by HAc writhing reaction,mice auricle swelling by induced xylene and hot plate method.The result showed that the total flavonoids could significantly inhibit the inflammation of mice auricles,alleviated pain of torsion reaction and increase pain threshold of hot-plate test mice.In addition,the analgesic effects of total flavonoids in processed drugs was greater than crude drugs.To study the antitumor activity of total flavonoids of processed and crude drugs in vitro and in vivo by MTT assay and model of H22 mouse.The research found that the total flavonoids of Diphylleia sinensis could inhibit the reproduction of A549 lung cancer cell and HepG2 cells.It also could inhibit the reproduction of cancer cells in mice.And the inhibition of processed drugs were greater than crude drugs.Using fluorescence staining with Hoechst33258 to test HepG2 cells after incubation,apoptotic morphology of was found HepG2 cells.To find the optimum extraction technology of polysaccharides from Diphylleia Sinensis,orthogonal experiment method was conducted and content was determined by UV.The optimum processing condition was that including extracted three times,each for 2 hours,temperature was 80 ℃ and the ratio of solid to liquid was 1:12.Gas chromatography was then used to analyze polysaccharides after purified.Result showed that the polysaccharides of Diphylleia Sinensis contained rhamnose,glucose,arabinose,galactose,xylose,mannose,ethylurethane,ribose,fructose.To study the anti-tumor activity of polysaccharides of Diphylleia Sinensis(PDS),it was not found that PDS could inhibit the reproduction of A549 and HepG2 cells in vitro.But it could inhibit the growth of H22 tumor in mice and increase the spleen index of tumor mice as well in vivo.To investigate the influence of polysaccharide on immune of mice by testing hemolytic,carbon granular clearancein,the ability of spleen cell transformation in mice and spleen lymphocyte proliferation in vitro.The results showed that the polysaccharides of Diphylleia Sinensis could increase the index of the clearance rate of carbon particles and restore immunity of micewith immune hypofunction.In addition,the hemolysin in serum and promote ability of spleen cell transformation in mice and the proliferation of spleen lymphocyte in vitro was improved.
