PART ONEObjective:To investigate the expression and their relationship of IncRNA-PVT1 and c-myc mRNA in human glioblastoma.Methods:Five cases of human glioblastoma tissue specimens and normal human glial tissue samples were obtained during operation.The expression of lncRNA-PVT1 and c-myc mRNA in human glioblastoma tissue specimens and normal human glial tissues were detected by agarose gel electrophoresis and fluorescence real-time quantitative PCR.Results:The expression of lncRNA-PVT1 was detected in human glioblastoma tissue specimens.The expression of lncRNA-PVT1 was not detected in normal human glial tissue samples.c-myc mRNA was expressed in glioblastoma and normal glial tissue,but the expression of c-myc mRNA was significantly increased in glioblastoma.There was no correlation between the expression of lncRNA-PVT1 and c-myc mRNA.Conclusion:The expression of lncRNA-PVT1 in human glioblastoma is abnormally high,and it is not expressed in normal human glial tissue.c-myc mRNA was expressed in glioblastoma and normal glial tissue,and the expression of c-myc mRNA in glioblastoma was significantly higher than that in normal glial tissue.The expression of lncRNA-PVT1 and c-myc mRNA in glioblastoma was abnormally increased,but there was no correlation between them.PART TWOObjective:To investigate the effect of lncRNA-PVT1 on the proliferation of human glioblastoma.Methods:The expression of lncRNA-PVT1 in U251 cells was reduced by small interfering RNA(siRNA)technique.The experiment was divided into NC group(siNC-U251 cells)and Si group(siPVTl-U251 cells).The expression of lncRNA-PVT1 and c-myc RNA were detected by qPCR.The cell proliferation was measured by CCK8 and EdU.The cell cycle was analyzed by flow cytometry.Western blot was used to detect the expression of c-myc protein and CyclinD1 protein in both groups.Results:The expression level of lncRNA-PVT1 was significantly lower than that of NC group,while the expression level of c-myc mRNA in Si group was not significantly different from that in NC group.CCK8 results showed that 1 day,2 days,3 days of cell viability Si group was significantly lower than NC group.EdU results showed cell proliferation ability,Si group was significantly lower than NC group.Cell cycle showed that Si group cells were blocked in G1 phase.Western blot showed that the expression of c-myc protein and CyclinDl protein in Si group was significantly lower than that in NC group(P<0.05).Conclusion:The expression of lncRNA-PVT1 in human glial cells decreased after transfection of siRNA,indicating successful transfection.c-myc mRNA levels were not significantly changed,indicating that lncRNA-PVT1 and c-myc mRNA in glioblastoma is not related,LncRNA-PVT1 had no effect on c-myc mRNA.LncRNA-PVT1 has a role in promoting the proliferation of human glioblast cells and can interfere with the process of human glial cell cycle.The expression of lncRNA-PVT1 is reduced and human glial cells are stagnated in G1 phase,The LncRNA-PVT1 may affect human glioblast proliferation by c-myc,CyclinDl and so on,and interfere with the cycle progression. |
