Objecti veThis paper aims to o investigate the therapeutic effect of Acid compound Rosmarinic acid extracted from Herba Sarcandrea on vitra pneumonia in mice caused by influenza H9N2.We also determined the mechanisms underlying the therapeutic effect of Rosmarinic acid on mice model of viral lung injury cause by H9N2 from the angles of anti-viral、anti-inflammatory and MAPK signaling pathway.Methods1.Effect and mechanism of Rosmarinic acid on viral pneumonia in mice caused by H9N2:Mice model was made by 2LD50 H9N2 by nasal drip.Rosmarinic acid was treaded after 2h infection.After 6 days of treatment,the lungs were removed and weighted.Lung index and Dry/wet ratio of lung were calculated.The morphological changes of lung tissue were observed and the viral load of lung tissue in each group was measured.2.Western blot analysis the expression of JNK、ERK、STAT3 and its phosphorylation of MAPK signaling pathway.3.mRNA assay of IL-1β、IL-6、RANTES、TNF of mice tissue by quantitative Real-time PCR.Results1.The Rosmarinic acid can effectively reduce the lung index and lung wet-dry-weight ratio of mouselet infected with H9N2 avian influenza virus(P<0.01,P<0.01);reduce the virus infect the virus capacity(P<0.01)of lung tissue of it and obviously mitigate lung tissue einjury by virus..2.When the mice was infected by H9N2 avian influenza virus,The level of phosphorylated protein(P<0.01,P<0.01,P<0.01)of JNK、ERK、STAT3 was significantly increased,the proteins family of MAPK signaling pathway,of the lung tissue of the mouselet.The Rosmarinic acid can obviously reduce the H9N2 avian influenza virus.to infect the level of phosphorylated protein(P<0.01,P<0.01,P<0.01)of JNK、ERK、STAT3,the proteins family of MAPK signaling pathway,of the lung tissue of the mouselet.3.When the mice was infected by H9N2 avian influenza virus,The level of mRNA(P<0.01,P<0.01,P<0.01,P<0.01)in the proinflammatory factor IL-1β、IL-6、RANTES、TNF of lung tissue of mouselet infected with H9N2 avian influenza virus was significantly increased.The level of mRNA(f<0.01,P<0.01,P<0.01,P<0.01)in the proinflammatory factor IL-1β、IL-6、RANTES、TNF of lung tissue of mouselet infected with H9N2 avian influenza virus was significantly decreased with the Rosmarinic acid with it.Conclusions1.The Rosmarinic acid can efficaciously restrain the inflammatory lung tissue of the mouselet being oozing by the H9N2,it can also restrain the duplication of influenza virus,relieve the damage of virus to lung tissue.2.The MAPK signaling pathway plays an important role in lung injury in mice after H9N2 avian influenza virus infection in mice.The Rosmarinic acid can effectively inhibit the level of phosphorylated protein of JNK,ERK,STAT3,the proteins family of MAPK signaling pathway,in order to regulate the expression of inflammatory factor,reduce the pathological damage of lung caused by H9N2 avian influenza virus.3.The Rosmarinic acid can effectively regulate the content of ERK signaling pathway downstream inflammatory factors IL-1,IL-6,TNF mRNA and JNK signaling pathway downstream chemokines;reduce the response of pulmonary inflammatory due to virus.
