Apigenin(API)is a widely found in fruits and vegetables of flavonoids,especially in the content of celery is high.In recent years,the pharmacological effects of apigenin have been studied at home and abroad,and it has been found to have anti-tumor,anti-inflammatory,oxidative damage protection,myocardial injury protection and lower blood pressure.3-chloro-1,2-propanediol(3-MCPD)is one of chloropropanol compounds,which is representative of the toxicity of chloropropanol compounds because of its high toxicity and high pollution.3-MCPD is often found in the acid-hydrolyzed plant protein(HVP)process.It has been found that 3-MCPD is more likely to be present in heated foods in the form of fatty acid esters.Excessive intake of these foods can results in its damage to the body,a serious cause of the body’s metabolic disorders.Our previous study found that exogenous apigenin can alleviate 3-MCPD-induced apoptosis and damage of human embryonic kidney cells.This experiment mainly from the body level to evaluate whether celery element can alleviate kidney damage caused by3-MCPD,by studying the mitochondrial function especially endogenous levels of oxidative phosphorylation and caspase apoptosis signaling pathways to explore celery element of renal injury caused by 3-MCPD protection mechanism.The main results were as follows:(1)Sprague Dawley(SD)rats were exposed to 28 days of intragastric administration by 3-MCPD doses of 15,30,60 mg / kg of low,medium and high doses.The results showed that: 3-MCPD can cause weight and food intake loss in rats,especially in the21 st day after high-dose oral administration.After 28 days of intragastric administration,the renal kidney was significantly higher than that of other treatments,especially in the high dose3-MCPD group,individual rat kidney appears whitening phenomenon;Further analysis of renal function showed that the blood urea and creatinine increased in the 3-MCPD treated group.Renal pathology biopsy showed that infected group processing ansing pathological condition,mainly displays in the renal edema,collecting duct epithelial cells arranged not neat,the cell body swelling,part of the cell necrosis,cytoplasm eosinophilic,hyperplasia ofrenal tubules,in many of the renal capsule.The results showed that 3-MCPD had obvious toxic effects on SD rats,which caused the damage of renal morphology and renal function in rats.kidney is the main target organ of 3-MCPD.(2)The 3-MCPD(30 mg/kg)and different doses of apigenin(20,40,80 mg/kg)in the coordination treatment for 28 days in rats.The results showed that: Apigenin can relieve3-MCPD-induced rat body weight and food intake,especially from the 21 st day,the high-dose apigenin group of food intake is higher than the exposure group;Apigenin can alleviate 3-MCPD-induced renal enlargement in rats;Further analysis of renal function found that apigenin to a certain extent,can reduce the blood glucose creatinine,urea content;Kidney pathology showed that compared with the 3-MCPD group,apigenin-treated nephropathy had a tendency to decrease.This shows that apigenin to a certain extent,can alleviate 3-MCPD caused by kidney damage.(3)The protective mechanism of apigenin on 3-MCPD-induced renal injury in rats was studied by mitochondrial function,especially oxidative phosphorylation and caspase endogenous apoptotic pathway.The results showed that: Apigenin can alleviate the decrease of mitochondrial membrane potential caused by 3-MCPD.Apigenin can alleviate the up-regulation of mitochondrial cytokine c protein in 3-MCPD-induced renal tissue Apigenin can up-regulate the gene and protein expression of mitochondrial respiratory chain complex II(SDHA),complex V(ATP6/ATP8);Apigenin can alleviate 3-MCPD-induced up-regulation of caspase-3 and caspase-9 at both gene and protein levels.This suggests that apigenin can alleviate 3-MCPD-induced renal injury in rats by regulating mitochondrial oxidative phosphorylation and caspase endogenous apoptotic signaling.
