| Gastrodiae Rhizoma is generated from the dry tuber of Gastrodia elata plants,which is a famous herbal grew in Sichuan.It is very effective in treating gout and spasm and in energizing body functions and relieving pain,and it is usually used to cure dizziness,limb paralysis,epilepsy,insomnia and high blood pressure,etc.What’s more,Gastrodiae Rhizoma is also applied in the area of health care,cosmetic,etc.,which is highly demanded in domestic and overseas market.Pingwu county of Sichuan province is the original producing area of Gastrodiae Rhizoma and local people already started to plant it in 1980s.At present,there are more than 1200 farmers households,and Gastrodiae Rhizoma standardized plants base in Pingwu county has already passed the national GAP certificate of SFDA.On the basis of the cross-fertilize quality issue and of the slow and outdated research and development problem,and with reference to study Sichuan Provinces special Project of enrich people and power county by technology,and the integration and industrialized development and demonstration of GAP plants craft in Pingwu county,this article is aimed at the quality evaluation and study of Gastrodiae Rhizoma cross-fertilize and its fine powder.Main study contents and results are as follows:1.On the basis of previous research,this article scientifically assessed the quality of cross-breed Gastrodiae Rhizoma from morphological histology,physical and chemical identification,examination and composition check,etc.,which aimed to provide scientific evidence for stable and controllable quality and for safe and useful clinical application of Gastrodiae Rhizoma.1.1 Morphological histology comparison of cross-fertilize Gastrodiae Rhizoma and its female and male parent plants to provided reference for cross-fertilize Gastrodiae Rhizoma identification and recognition.When it came to the modality of original plant,we could distinguish it from plants height,stem and blossom color,and inflorescence length,etc.,which was different from cross-fertilize one.When it came to medicine character,we could distinguish it from its tuber shape,size and surface characteristics,etc.When it came to microscopic characteristics,for the first time we analyzed such aspects under microscope as needle crystal beam,vessel and polysaccharide particles of cross-fertilize and its female and male parent plants.The results showed that there was difference between the size of above-mentioned items of cross-fertilize and its female and male parent plants.1.2 Thin-layer chromatography(TLC)and HPLC fingerprint comparison of cross-fertilize Gastrodiae Rhizoma and its female and male parent plants.After TLC comparison and analysis,we found there was a little difference between TLC spot size and color depth of cross-fertilize Gastrodiae Rhizoma and its female and male parent plants even though it was same color chromatographic spots on the corresponding position with substance and contrast medicinal.which indicated that the content of chemical composition was slightly different between them.According to the comparison of HPLC fingerprint.the number of chromatographic peaks was the same as each other,but peak area was different.which proved that chemical composition was same,but amount was different.1.3 In reference to Chinese Pharmacopoeia version 2010,we checked and estimated different samples’ content of moisture,total ash,sulphur dioxide and extracts,and found that the lowest ratio was respetively 14.1%,4.0%.2.72 mg/kg and 16.0%,and the highest ratio was respectively 9.0%,2.1%;1.14 mg/kg and 7.2%.that sulphur dioxide content of all samples confirmed with the regulations in Chinese Pharmacopoei.and that extracts content of three batches of samples didn’t.two of them were Gastrodia elata Bl.f.elata × Gastrodia elata Bl.f.glauca S.Chow..and one of them was astrodia elata Bl.f.glauca S.Chow × Gastrodia edata Bl.f.elata.1.4 On the basis of previous research,we have measured the content of gastrodin and polysaccharides.Besides,we also analyzed the important data which was related to the content of extracts.gastrodin and polysaccharides with comprehensive evaluation method,which showed that the content of them from highest to lowest was Gastrodia elata Bl.f.glauca S.Chow × Gastrodia elata Bl.f.elata.Gastrodia elata Bl.f.glauca S.Chow、Gastrodia elata Bl.f.elata and Gastrodia elata Bl.f.elata ×Gastrodia elata Bl.f.glauca S.Chow.2.Researched on gastrodia ultra-fine powders from the quality,efficacy and safety,dissolution rate of different fineness to provided scientific evidence to its quality control and clinical application.2.1 Firstly,we prepared four specifications of ultra micro powder,including 40μm、20 μm、10 μm and 5 μm.Then we compared them with normal powder from the aspects of the characters,microscopic characteristics and TLC identification.On the aspect of ultra micro powder characters,all of them were yellowish-white,tiny smell,sweet taste,but the powder was finer and easier gathered than normal one.There were no complete cells under ordinary biology microscope,only cell debris.We could spot gastrodin by detecting TLC,and there were same chromatographic spots in related position of medicinal sample.2.2 By checking different specifications of gastrodia ultra micro powder,from the aspects of routine inspection.SO2 residue,heavy metals and harmful elements,we found that the highest of moisture content was 7.1%the lowest was 6.8%,the total ash upped to 2.45%,the minimum was 2.28%,both of them were lower than normal powder:acid insoluble ash upped to 0.05%,the minimum was 0.02%,SO2 residue upped to 2.08 mg/kg.the minimum was 1.79 mg/kg.there was no difference between both of them compared with the ordinary powder;heavy metals,harmful elements content and microbial limit inspection were in accordance with the stipulations in the 2015 edition of Chinese Pharmacopoeia limit value and oral medicine preparations containing medicinal powder preparation requirements,respectively.2.3 By measuring bulk density and repose angle two indicators to evaluate the liquidity of the different specifications of gastrodia ultra micro powder and ordinary powder,the results showed that about the liquidity there were no differences between different specifications of gastrodia ultra micro powder,but all of them were lower than the ordinary powder:compared with ordinary powder,the bulk density of ultra micro powder was increased.it indicated the quality of unit volume ultra micro powder was larger.2.4 Having a assaying of extract and gastrodin content of different specifications of gastrodia ultra micro powder and ordinary powder,it showed that the extracts and gastrodin content of ultra micro powder was higher than ordinary powder.the extract of 10 μm was higher,the gastrodin content of 20 μm was higher.2.5 For the first time,simulating the conditions in human body,studying on the gastrodin dissolution rate of different specifications of gastrodia ultra micro powder and ordinary powder,the study showed that after superfine grinding processing.the dissolution of gastrodin was increased,and that of the 20μm was the highest.2.6 By researching on safety evaluation,anticonvulsive and sedation for the first time,provided scientific evidence for clinical application of gastrodia ultra micro powder.The results of acute toxicity test showed that the maximum tolerance of mice to different specifications of gastrodia ultra micro powder and of ordinary powder was more than 15 g/kg.which was 150 times as adult daily doses tolerance,so non-toxic.The results proved it was safe.Using PTZ to cause convulsing in mice as the model,we compared the effect of treating spasm of different specifications of ultra micro powder and normal powder.it showed that different specifications of ultra micro powder and ordinary powder had significant differences,the anticonvulsive effect of the superfine powder was significantly enhanced(p<0.05).and the effect of 20 μm was the most useful.Using Chloral hydrate to make mice sleeping as the model.and studying the sedation effect of different specifications of ultra micro powder and normal powder.it showed that the different specifications of ultra micro powder and ordinary powder compared with samples had significant differences(p<0.05).the superfine powder can prolong sleeping time of mice.the sedation effect was enhanced(p<0.05),and the effect of 20 μm was most useful.To sum up,from the aspects of medicinal usage.compared with its female parent and male parent,the quality of Gastrodia elata Bl.f.glauca S.Chow×Gastrodia elata Bl.f.elata was better,the quality of Gastrodia elata Bl.f.elata × Gastrocdia elata Bl.f.glauca S.Chow was poor.From the aspects of quality,absorption,the efficacy and safety,and considering the actual production.the 20 μm was the best. |
