| Skin whitening and lightning has become an important direction of research because of the pursuit of white skin.Hormone and hydroquinone used in mordern western society,despite high-effciency,but these compounds are residual in vivo.Triacid,vitamin C which added to cosmetics have the weakeness of low stability or slow performance.TCM,with its advantages of safety,mildness,low toxicity,high-effciency,is becoming research focus.This kind of resource is abundant in our hometown,what’s more is that our ancestors have studied this for over 2000 years.The root of Coptis chinensis Franch,as an important traditional Chinese medicine,was usually digged out from the 5th year.However,C.chinensis inflorescence,as a by-product,is often disposed as waste.This not only wasted the resources,but also consumed the nutrition as well as affects the quality of rhizome.So,it is essential to remove the inflorescence at the beginning of flowering phase.Nowadays,people at home are mainly children,elders and women.For the situation,the enthusisasm for picking up the inflorescence was decreased.Most inflorescence bloomed and then falled.We deduce that such condition may be improved by some valued products of C.chinensis inflorescence.If we can make full of C.chinensis inflorescence and increase the market demand,the enthusiasm could be aroused and prevent the waste of C.chinensis inflorescence resource.So we think if we could combine the study spot with the use of C.chinensis inflorescence?Therefore we study the C.chinensis inflorescence and linarin whcih was first founded from C.chinensis inflorescence of skin-whitening and lightning.We got this results:(1)We detect the content of linarin in the water extract of C.chinensis inflorescence,it’s 22.31±1.18 mg/g.Then we used three different methods to study C.chinensis inflorescence extract and antioxidant activity of flower glycosides,the results was given that C.chinensis inflorescence extract has certain DPPH and ABTS radical scavenging ability and reducing power;The DPPH radical scavenging ability and reducing power of linarin was weaker,while has certain ABTS radical scavenging ability.(2)We studied the effects of linarin and C.chinensis inflorescenc in vitro on tyrosinase,the results found that linarin in low concentration range(25-100μg/ml),its effect of inhibiting monophenoloxidase tyrosinase enzyme activity is better than thatof two kinds of positive control,in the 200 μg weaker than arbutin;And C.chinensis inflorescenc in low concentration range(25-100μg/ml),its effect of inhibiting monophenoloxidase tyrosinase enzyme activity is weaker than both positive control,stronger than the nicotinamide on 200 μg/ml.The lianrin’s inhibition effect of diphenolase tyrosinase was better than positive in these five concentrations.At low concentration of C.chinensis inflorescenc,its effect of inhibiting diphenolase tyrosine is weaker than positive control,however,when the concentration of 166.67 μg/ml its effect is better than that of the two kinds of positive control.Under the conditions of 166.67 μg/ml,the inhibition rate of the C.chinensis inflorescence and linarin was 26.18%and 34.24%respectively.(3)By ultraviolet spectrum research,we received the effect of samples on dopamine oxidation product and copper ions,the results showed that linarin and C.chinensis inflorescenc can inhibit melanin by reacting with dopamine quinone formation.C.chinensis inflorescenc.The C.chinensis inflorescenc doesn’t have any chelation with the copper ions,but it produces something that makes the ultraviolet absorption increase.Linarin is not only chelating with the copper ions,but also produces something that makes the ultraviolet absorption increase.Linarin can be speculated to the reason of inhibiting the tyroniase activity is that 4’-hydroxyl group in C ring which may interactes with tyroniase.(4)By fluorescence spectra,it is found that both linarin and C.chinensis inflorescence can reduce the characteristic fluorescence,and the linarin is static quenching.the absorption peaks after the addition of copper ions in the linarin solution changes from 320nm to 323nm.As flavonoid,linarin can be speculated to the reason of inhibiting the tyroniase activity is that 4’-hydroxyl group in C ring which may interactes with tyroniase.(5)From the cellular level,by MTT experiment,inhibiting tyrosinase activity illustrate the mechanism of C.chinensis and linarin affecting the tyrosinase.The result shows that both of them can achieve the aim of skin-whitening through inhibiting the B16 cell preliferation and activity of tyrosinase.In sum,C.chinensis.and linarin both have the certain ability to inhibiting the activity of tyroniase.In this paper we explored the mechanism of inhibiting the tyroniase activity briefly.Specially,linarin has the ability to be comparable to the positive control,which has the potenty to study further. |
