Beta-eleostearic acid,one isomer of conjugated linolenic acids with three all-trans-conjugated double bonds,has been reported to exert anti-tumer and anti-metastatic effects.However,there is no report on the inhibition of β-eleostearic acid to choriocarcinoma.In this study,MTT assay was performed to reveal that β-eleostearic acid can inhibit the proliferation of choriocarcinoma JEG-3 cells in dose-and time-dependent manners.Afterwards we observed the morphology changes of JEG-3 cells via Hoechst 33342 fluorescence staining assay.Staining with Annexin V/PI,JEG-3 cells were determined by flow cytometer,which demonstrated that p-eleostearic acid could induce JEG-3 cells apoptosis.In addition,through Western Blot we proved that the apoptotic mechanism caused activation of caspase-3 and caspase-9 as well as down-regulation of the ratio of Bcl-2/Bax.Besides,using the specific inhibitors,we found that blocking p38MAPK and PPARγwould restrain cell apoptosis.This further indicated that JEG-3 cells were induced apoptosis by β-eleostearic acid through the continuous activation of p38MAPK,PPARγ as well as p53.Wound-healing assay indicated that β-eleostearic acid might hinder the migration of JEG-3 cells,and the invasion assay confirmed its anti-invasion effect.Furthermore,gelatin zymography showed that β-eleostearic acid suppressed the migration of JEG-3 cells as a result of suppressing the secretion of MMP-2 and MMP-9.These results suggested that β-eleostearic acid induced the apoptosis and suppressed the migration of JEG-3 cells.The effect of apoptosis induction involed the p38MAPK activation and PPARγ protein expression regulation.
