| Contrast agents are the commonly used drugs in modern radiology,which are mainly used in angiography and endoradiography.However,as for the characteristics of high osmosi,high vicidity,low electronegativity and chemical toxicity,the administration of contrast agents can cause adverse effects to body function and hemorheology,sometimes may even lead to kidney failure(KF).Objective:The present study is to find the effects of non-ionic contrast agent on the hemorheology both in vivo and in vitro.Methods:Human peripheral blood samples were collected in tubes at the time of 30 min,1h,and 2h after adding contrast agents,and whole blood viscosity(HV),hematocrit(Hct),erythrocyte deformation index(DI),erythrocyte electrophoresis rate and erythrocyte osmotic fragility were measured.Rabbit blood samples were collected in tubes at the time of 30 min,2h,6h and 24h after administration of contrast agents,and all the indexes above including the(DI)d and the orientation index[(DI)or]were measured.Results:1.In vitro,the whole blood viscosity(HV)at different shear rates increased significantly(p<0.01)at the time of 1 h after adding contrast agents;The erythrocyte electrophoresis rate decreased at the time of 30 min and 1 h(p<0.05);Although the erythrocyte deformation index(DI)slightly decreased and the erythrocyte osmotic fragility slightly increased,both of which had no statistical significance.2.In vivo,however,the whole blood viscosity(HV)at different shear rates decreased(p<0.05)at the time of 6h after administration of contrast agents;The erythrocyte deformation index(DI)decreased at the time of 30min(p<0.05);The(DI)d decreased at the time of 2h(p<0.05),while the orientation index[(DI)or]decreased at the time of 30 min,2 h and 6h(p<0.05).Although the erythrocyte osmotic fragility increased,it had no statistical significance.Conclusion:The administration of contrast agents could dilute the blood and decrease the hematocrit(Hct),while the low electronegativity and chemical toxicity of the contrast agents may lead to hemolysis.The(DI)d indicate the fluxility of erythrocyte membrane.The decreased(DI)d may due to the inreversible harm caused by oxidative stress.Although the blood volume increased,the effective circulating blood volume(ECBV)decreased on the contrary,this may partly lead to the decrease of the whole blood viscosity(HV).The hardening of the red blood cells(RBCs)decreased their plasticity,which may be involved in the decrease of the erythrocyte deformation index(DI).The adverse change of the glomerular filtration rate(GFR),Erythrocyte surface charge,Erythrocyte aggregation,the red blood cell deformability and the endothelial cell adhesion lead to the aggregation of the red blood cells(RBCs),increased the resistance of blood flow and decreased the effective circulating blood volume(ECBV),which may worsen the anoxia and hypoxia of renal medulla and cause mechanical damage to renal medulla. |
