The Role Study Of Astragalus Leech Preparation On Expression Of C-Ⅳ And TGF-β1 Of Renal Tissue In STZ-induced Diabetic Nephropathy Rats

Objective To investigate the effect of Astragalus leech preparation on expression of An egg reduce and Kidneys protection in STZ-induced diabetic nephropathy rats,and further explore the mechanism of action of Astragalus leech preparation on the renal protection in STZ-induced Diabetic Nephropathy RatsMethods In animal experiments,using male SD rats,high fat,high-calorie diet-induced and repeated intraperitoneal injection of streptozotocin diabetic rats were established.The last two weeks after STZ injection,urine MA/Cr,urine MA/Cr>30mg/g for the successful modeling;A total of 32.randomly divided into four control groups:model control group,Chinese medicine group(Astragalus leech preparation group),western medicine group(Lotensin group)and Chinese and Western medicine group(Astragalus leech preparation and Lotensin group),at the same time,set up the normal control group.Take appropriate intervention for 8 weeks after detection of the rats body weight(BW),kidney weight index,fasting blood glucose(FBG),glycosylated hemoglobin,24h urinary albumin excretion rate(UAER),urinary al-microglobulin(al-MG),urinary β2-microglobulin(β2-MG),urinary retinol binding protein(RBP)and transferring(TRF)and immunohistochemical detection of large rat kidney C-IV and TGF-β1,improve hemodynamics play efectResults Animal experiments,the general situation in all treatment groups,biochemical indicators,changes in renal indices were better than the model group,while C-Ⅳ and TGF-β1,expression was significantly reduced,renal pathological change mitigation,in which Astragalus leech preparation treatment group,Astragalus leech preparation and and Lotensin group improved most significantly(P<0.05),two groups showed no differenceConclusion Clinical syndromes of Astragalus leech preparation formula can obviously improve the diabetic nephropathy,and can reduce urinary albumin excretion.The possible mechanism of the renal protective effect of Astragalus leech preparation on STZ-induced Diabetic Nephropathy Rats was the inhibited expression of C-IV and TGF-β1 in renal tissue in rats.To improve the high renal blood flow,high perfusion,inhibit the early stage of renal hypertrophy,reduce proteinuria and protect renal function,and delay the process of renal fibrosis.
