Objective Myocardial injury is a common complication of endotoxemia,the incidence of about 50%,has high mortality.This experiment by tail vein injection of lipopolysaccharide(LPS)solution,to make the Preparation of endotoxemia myocardial injury model in rats.To study of dehydrogenation corydaline(DHC)on rats myocardium under endotoxemia bypass on its Protective effect and mechanism.Methods 50 male Wistar rats were randomly divided into five groups:NS control group.LPS model group,DHC low dose group,DHC medium dose group and DHC high dose group.Each groupof 10.Through the tail vein injection of lipopolysaccharide(LPS)to establish endotoxemia model of myocardial injury,the concentration of 5 mg/Kg.LPS model group and DHC intervention group injected lipopolysaccharide,NS control group injected saline volume,etc.Then to intervene these models by low(2.5mg/Kg),medium(5mg/Kg)and high(10mg/Kg)doses of DHC,NS control group and LPS model group injected saline volume,etc.At 1 hour after the medication,take blood and the myocardial tissue,CK enzyme,CKMB serum enzyme and cTnI levels were measured by enzyme rate method,the TNF-a,IL-1β levels were assayed by ELISA method,Myocardial tissue using HE staining to Pathology observation,and using the immunohistochemical SP method to detect myocardial tissue TNF-alpha,IL-1beta.Results 1 Pathological observation showed that edema,inflammatory cells infiltration and necrosis foci in LPS model group,but the injury was significantly alleviated in DHC low and medium dose group,then DHC high does group was between them.2 Serum CK,CK-MB,Tnl content determination results showed that compared with the NS control group,CK,CKMB and cTnI levels of LPS model group were increased remarkably(P<0.05).Meanwhile,the levels of low and medium dose groups were decreased significantly.But there were no significantly different in the high dose group.3 The TNF-a and IL-1β detection results showed that compared with the NS control group,LPS model group were decreased significantly(P<0.01),DHC with low,medium and high dose groups compared with LPS model group significantly reduced(P<0.01).4 Immune histochemical method in the determination of myocardial tissue TNF-a,IL-1β the results showed that:LPS model group compared with NS control group,TNF-a,IL-1βcontent increased significantly,the difference was statistically significant(P<0.01);DHC with low,medium and high dose groups compared with LPS model group were decreased significantly,the difference was statistically significant(P<0.05).5 Immune histochemical method in the determination of myocardial tissue NF-κB the results showed that:compared with NS control group,LPS model group increased significantly,the difference was statistically significant(P<0.01);DHC with low,medium and high dose groups compared with LPS model group were decreased significantly,the difference was statistically significant(P<0.01).Conclusion This study observed the earliest damage of endotoxin to rat myocardium appeared after lhour of endotoxin injection.The dehydrogenation corydaline could inhibit inhibit the NF-kappa B expression,reduce the generation of TNF-alpha,IL-1 beta,reduce myocardial injury effectively,especially in low and medium groups.
