The Correlation Of Adenovirus Type 36 Infection And Adipokine Adiponectin Expression | Posted on:2015-04-01 | Degree:Master | Type:Thesis | Country:China | Candidate:X Chang | Full Text:PDF | GTID:2404330488999034 | Subject:Biochemistry and Molecular Biology | Abstract/Summary: | PDF Full Text Request | Objective:1)To detect the the antibody of Ad36 and examine the serum adiponectin and the expression level of adiponectin mRNA in abdominal omental,subcutaneous adipose tissues and according to Ad36 infection or not,analysis its relationship with the factors of fasting glucose and lipids,etc.To investigate the correlation of adenovirus type 36 infection and adiponectin expression in Uygur obese patients.2)3T3-L1 preadipocyte cell lines as an vitro experimental model by examining adiponectin gene expression during the period of 3T3-L1 preadipocyte differentiation,to further explore the regulation of Ad36 infection on adiponectin expression.Methods:According to the diagnostic criteria of obesity,the samples were divided into obese group and non-obese group.1)Using serum neutralization test to detect the antibody of Ad36.2)Using the ELISA method for the determination of serμm adiponectin protein levels.RT-PCR was used to investigate the expression of adiponectin mRNA from SAT and OAT.3)3T3-L1 preadipocyte were cultured invitro and with different MOI of Ad36 infected it before its induced to differentiated into the matured adipocytes.The differentiation of adipocytes was assessed through oil red O staining and detecting expression of adiponectin mRNA by real time RT-PCR according to Ad36 infection at different times(0-7days).Results:1)Compared with the non-obese group,obese patients infected with Ad36 were significantly higher than that of non-obese group(P<0.05).2)Serum adiponectin was significantly reduced in obese group than non-obese group(P<0.05).In obese group,Serum adiponectin was significantly increased in the Ad36-infected group than non-infected group(P<0.05)and the cholesterol(TC)and triglyceride(TG)levels have a downward trend.3)In the fifth day and seventh day of Ad36 infection with 2 MOI(3.8and7.6),the level of adiponectin gene mRNA expression was obviously up-regulated than the non-infected day(P<0.01).Conclusion:1)Ad36 infection may be associated with the occurrence of Uygur obesity.2)Adenovirus type 36 infection up-regulate the expression of adiponectin levels. | Keywords/Search Tags: | Ad36, Adiponectin, Obesity, 3T3-L1 adipocyte, Real time RT-PCR | PDF Full Text Request | Related items |
