Ornithogalum caudatum Ait(OCA)is a kind of perennial herb belongs to liliaceae ornithogalum linn,with the effect of clearing heat and detoxicating,resolving hard lump.It can treat innominate inflammatory,hepatitis,liver cirrhosis and liver cancer,etc.Compound Wannianqing Capsules which take OCA as the main herb has great effect on treating lung cancer,gastric cancer and liver cancer.According to the literature published,OCA polysaccharide has great antineoplastic activity with a high content in OCA.However,most of the research of OCA only used simple extract,and focus on the optimization of extraction process,as well as the effect on tumor-bearing mice’s immune system.The inhibiting effect of OCAP on tumor cell proliferation in vivo and in vitro is also studied.But rarely has research focus on the modification of structure and structural features.Researches show that Se-polysaccharide has the combined activity of selenium and polysaccharide,higher than the single use.It becomes the popular research project as a new kind of antitumor drug.Thus the research combines OCAP with sodium selenite to compound Se-polysaccharide,modify the se-rich technology.Observe the physicochemical property and structure characteristics,explore the effect on antioxidant activity of H22 tumor-bearing mice.Firstly,prepare OCAP and modify the selenizing technology.The materials are extracted by ethanol.By using alcohol sedimentation method,crude polysaccharide can be extracted from OCA.Protein is removed by Papain-Sevag method.Pigment is removed by hydrogen peroxide solution.Fractional step alcohol sedimentation method and DEAE-52 column chromatography are used to purify OCAP.Finally the optimal composition of OCAPIIA will achieve.Selenium content is taken as index,response surface methodology is used to select optimal selenizing technology.The optimum includes response time:8.09h;reaction temperature:71℃;mass ratio of Na2SeO3 and OCAPIIA:0.97g/g.Under this condition,selenium content of OCAP is 3.137mg/g.Secondly,physicochemical property and structure characteristics have been observed by phonel-sulfate method,GC analysis and scanning electron microscope.The results show that Se-OCAPIIA is a kind of solid white flocculent powder,tasteless,soluble in water and saline,soluble in acid and alkali solution,insoluble in organic solvent.It turns reddish-brown colour in pheno-sulfuric acid reaction,indicates that it contains polysaccharide.The morphology is different from Se-OCAPIIA and OCAPIIA,OCAPIIA presents banding,blade shape and strip,while Se-OCAPIIA mainly presents globular and cotton-shape.Monosaccharide composition and molar ratio of Se-OCAPIIA point that D-glucose:D-mannose:D-galactose=17:4:10.In the end,effect on antioxidant activity of H22 tumor-bearing mice has been observed.The results indicate that Se-OCAPII can significantly elevate the activity of catalase(CAT),superoxide dismutase(SOD)in liver and serum glutathion peroxidase(GSH-Px).On the contrary,MDA is decreasing in H22 tumor-bearing mice.Se-OCAPII strengthens the ability of disintegrating oxygen free radicals that improve antioxidant activity of H22 tumor-bearing mice.Under the same dose,Se-OCAPII is superior to OCAPII in promoting the expression of CAT,SOD and GSH-Px,as well as reducing the expression of MDA.But the difference has no statistical significance.Tumor inhibitory rate and immune organ index have also been studied.The results indicate that Se-OCAPII intraperitoneal injected with 50,100,200mg/(kg*d)respectively,the tumor inhibitory rate is 32.36%,57.77%and 66.24%.The weights of tumor decline compared with control group.Under the same dose,Se-OCAPII is superior to OCAPII in inhibiting the tumor growth,but the difference has no statistical significance.Spleen index and thymus index increase by the use of Se-OCAPII.Under the dose of 100-200 mg/(kg d),thymus index is significantly higher than the control group(P<0.01).Under the same dose,Se-OCAPII is superior to OCAPII in increasing spleen and thymus index,but the difference has no statistical significance.To sum up,the research confirms that optimal response time is 8.09h,temperature is 71℃,mass ratio of Na2Se03 and OCAPIIA is 0.97g/g,selenium content is 3.137mg/g of the best selenizing technology.The monosaccharide composition of Se-OCAPIIA indicates D-mannose:D-glucose:D-galactose=17:10:4,with the morphology of globular and cotton-shape.Se-OCAP has great antioxidant and anticancer effect on H22 tumor-bearing mice,which is superior to OCAP.The research makes a foundation for the further development and utilization of OCAP.
