| Purpose Through assay expression changes of Bax、Bcl-2 in thyroid gland tissues with different blood glucose concentration in DM state,to approach injury degree and possible mechanism of different blood glucose concentration to thyroid gland function in DM rats.Methods Replicationed DM rats.divided the DM rats to persistence hyperglycemia group(N=10)、blood glucose fluctuation group(N=10)and control group.After 12 weeks.executed the rats,weighed bilateral thyroid gland,accounted thyroid gland weight index.Use immunohistochemistry assayed expression of Bcl-2,Bax and albumen,scanning electron microscope observed ultra-structure,flow cytometry assayed apoptotic rate in thyroid gland tissues.Results 1.The thyroid gland weight in diabetes rats which had syndromes of intermin-gled phlegm and blood stasis was higher than group N.P>0.05;The thyroid gland weight index in diabetes rats which had syndromes of intermin-gled phlegm and blood stasis was obviously lower than group N(P<0.01).There was no significance difference between persistence hyperglycemia group and blood glucose fluctuation group.2.Immunohistochemistry assay:Under light microscope.we observed HE staining,finding that karyotin of thyroid gland follicular epithelial cell in group N was minus,while in diabetes rats which had syndromes of intermin-gled phlegm and blood stasis,it was minus or weakly positive,more than group N.There was no significance difference between persistence hyperglycemia group and blood glucose fluctuation group.Figure assay results:Immunohistochemistry staining positive area ratio and average opticaldensity of thyroid gland tissues in diabetes rats which had syndromes of intermin-gled phlegm and blood stasis,were higher than group N;Bax staining positive area ratio of DM rats was bigger than group N,however,there was no statistics difference between two groups of Bax staining average opticaldensity.Bcl-2 immunohistochemistry staining positive area ratio and average opticaldensity of diabetes rats which had syndromes of intermin-gled phlegm and blood stasis were lower than group N(P<0.01).Bcl-2/Bax positive area ratio and average opticaldensity ratio of diabetes rats which had syndromes of intermin-gled phlegm and blood stasis were lower than group N(P<0.01).There was no significance difference between persistence hyperglycemia group and blood glucose fluctuation group.3.Biochemistry index assay:Compared with group N,CHOL and FPG and HbA1c of diabetes rats which had syndromes of intermin-gled phlegm and blood stasis were obviously higher than the normal group,differences had statistics significance;TRIG in persistence hyperglycemia group was obviously higher than the normal group(P<0.01).differences had statistics significance;FPG and CHOL in blood glucose fluctuation group were obviously lower than persistence hyperglycemia group.differences had statistics significance;TRIG in blood glucose fluctuation group was obviously lower than persistence hyperglycemia group(P<0.01),differences had statistics significance;HbA1c in persistence hyperglycemia group was close to blood glucose fluctuation group,differences had no statistics significance.4.Insulin assay:Hyperinsulinism occured in diabetes rats model which had syndromes of intermin-gled phlegm and blood stasis,both in persistence hyperglycemia group and blood glucose fluctuation group,and there were obvious differences between two model groups and the normal group,also between the two model groups.5.Blood glucose fluctuation index assay Compared with group N,MBG and LAGE and SDBG of diabetes rats which had syndromes of intermin-gled phlegm and blood stasis were obviously higher than group N,differences had significance;MBG and LAGE and SDBG of blood glucose fluctuation group were obvious differences between two model groups.6.Thyroid gland function assay:Freeing T3 and freeing T4 of in diabetes rats model which had syndromes of intermin-gled phlegm and blood stasis,both in persistence hyperglycemia group and blood glucose fluctuation group,were obviously lower than the normal group,and differences had significance.There was no significance difference between persistence hyperglycemia group and blood glucose fluctuation group.There was no significance difference between TSH of every two groups.7.Thyroid gland ultra-structure assay:After 12 weeks,there was no obvious pathology change in thyroid gland tissues of rats in the normal group,but follicular epithelial cells of diabetes rats in the group which had syndromes of intermin-gled phlegm and blood stasis,mostly showed thin and flat,lost microvilli.There was no significance difference between persistence hyperglycemia group and blood glucose fluctuation group.Conclusion 1.Diabetes rat model which had syndromes of intermin-gled phlegm and blood stasis could be successfully maked by high glucose and fat diet feeding combine with low dose STZ caudal vein injection.2.Low thyroid gland function in diabetes rats which had syndromes of intermin-gled phlegm and blood stasis.ultra-structure changes of thyroid gland tissues hinted that injuries occured in thyroid gland follicular epithelial cells.Increase Bax expression of thyroid gland epithelial cells,decrease Bcl-2 expression,obvious Bcl-2/Bax descent,increase thyroid gland follicular epithelial cells apoptosis under DM state,were likely to be one of thyroid gland injury mechanisms.But there was no significance difference between persistence hyperglycemia group and blood glucose fluctuation group,which explained that blood glucose fluctuation had no obvious injury to thyroid gland tissues in diabetes mellitus state. |
